以非洲山毛豆脱脂粉末为原料制备富含多肽的酶解液,并对其抗氧化活性进行了研究.以水解度和清除1,1-二苯基-2-三硝基苯肼(1,1-Diphenyl-2-picrylhydrazyl radical 2,2-Diphenyl-1-(2,4,6-trinitrophenyl)hydrazyl,DP-PH)自由基为指标,选择最佳水解酶,在酶解时间、温度、pH、酶与底物比例、底物与水比例等单因素试验基础上,利用正交试验对最佳的酶解工艺条件进行优化,同时对酶解产物对DPPH.、.OH和O2-.的清除能力和抗脂质过氧化反应能力进行了研究.结果表明,碱性蛋白酶为最佳水解酶,其酶解最佳条件为:50℃pH 8.0、m(酶)∶m(底物)=2∶100、m(底物)∶m(水)=5∶100,酶解4 h;酶解液清除自由基能力大小顺序为DP-PH.〉.OH〉O2-.和抗脂质过氧化作用,最高清除率分别是91.58%、90.19%、43.56%和44.73%;当质量浓度大于0.6 g/L,酶解液抗脂质过氧化作用高于2,6-二叔丁基-4-甲基苯酚(Butylated hydroxytoluene,BHT).

Defatted powder of Tephrosia vogelii was hydrolyzed to prepare hydrolysate rich in peptides,and its antioxidant activity was investigated by free radical scavenging assay.The best hydrolysis enzyme was selected on the degree of hydrolysis and the abilities to scavenge 1,1-Diphenyl-2-picrylhydrazyl radical 2,2-Diphenyl-1-(2,4,6-trinitrophenyl)hydrazyl(DPPH)free radical.Further,single factor and orthogonal experiments was employed to optimize pH,time,temperature,enzyme-to-substrate ratio and substrate-to-water ratio for maximizing polypeptide content.Besides,the scavenging activity of DPPH ·,· OH and O2-·and anti-lipid peroxidation were studied.The results showed that the most suitable enzyme was alcalase and the optimal hydrolysis conditions were 50 ℃ pH 8.0,m(substrate)∶ m(water) =5∶ 100,m(enzyme)∶ m(substrate) =2∶ 100,hydrolyzing 4 h.The effect on free radical scavenging capacity of Tephrosia vogelii polypeptides in the order was DPPH · 〉 · OH〉O2-· and liposome peroxide,the highest clearance rate were 91.58%,90.19%,43.56% and 44.73%,respectively.The polypeptide had strong anti-lipid peroxidation effect,higher than butylated hydroxytoluene(BHT),when its mass concentration were greater than 0.6 g/L.
