目的:观察乳化氟碳保存液和高渗枸橼酸盐嘌呤溶液对低温保存肾脏抗氧化能力和超微结构的影响,探讨乳化氟碳保存液在供体肾脏保存中的作用。方法:选择24只雄性SD大鼠随机分为高渗枸橼酸盐嘌呤溶液组(HCA)和乳化氟碳液组,每组12只。分别采用乳化氟碳保存液和高渗枸橼酸盐嘌呤溶液原位灌注双侧肾脏后快速切取,置于0~4℃20 mL上述相应液体中保存24 h后,比较两组保存液对大鼠肾脏的肾皮质匀浆MDA含量、SOD活性及Bcl-2基因表达和形态学改变。结果:乳化氟碳液组肾皮质匀浆MDA含量明显低于HCA液组(P<0.05),SOD活性、Bcl-2基因表达明显高于HCA液组(P<0.05),病理评分低于HCA液组(P<0.05),肾小管上皮细胞损伤轻于HCA液组。结论:乳化氟碳液可提高供体肾抗氧化和抗凋亡能力,减轻供体肾保存期的肾损伤。
目的:觀察乳化氟碳保存液和高滲枸櫞痠鹽嘌呤溶液對低溫保存腎髒抗氧化能力和超微結構的影響,探討乳化氟碳保存液在供體腎髒保存中的作用。方法:選擇24隻雄性SD大鼠隨機分為高滲枸櫞痠鹽嘌呤溶液組(HCA)和乳化氟碳液組,每組12隻。分彆採用乳化氟碳保存液和高滲枸櫞痠鹽嘌呤溶液原位灌註雙側腎髒後快速切取,置于0~4℃20 mL上述相應液體中保存24 h後,比較兩組保存液對大鼠腎髒的腎皮質勻漿MDA含量、SOD活性及Bcl-2基因錶達和形態學改變。結果:乳化氟碳液組腎皮質勻漿MDA含量明顯低于HCA液組(P<0.05),SOD活性、Bcl-2基因錶達明顯高于HCA液組(P<0.05),病理評分低于HCA液組(P<0.05),腎小管上皮細胞損傷輕于HCA液組。結論:乳化氟碳液可提高供體腎抗氧化和抗凋亡能力,減輕供體腎保存期的腎損傷。
목적:관찰유화불탄보존액화고삼구연산염표령용액대저온보존신장항양화능력화초미결구적영향,탐토유화불탄보존액재공체신장보존중적작용。방법:선택24지웅성SD대서수궤분위고삼구연산염표령용액조(HCA)화유화불탄액조,매조12지。분별채용유화불탄보존액화고삼구연산염표령용액원위관주쌍측신장후쾌속절취,치우0~4℃20 mL상술상응액체중보존24 h후,비교량조보존액대대서신장적신피질균장MDA함량、SOD활성급Bcl-2기인표체화형태학개변。결과:유화불탄액조신피질균장MDA함량명현저우HCA액조(P<0.05),SOD활성、Bcl-2기인표체명현고우HCA액조(P<0.05),병리평분저우HCA액조(P<0.05),신소관상피세포손상경우HCA액조。결론:유화불탄액가제고공체신항양화화항조망능력,감경공체신보존기적신손상。
Aim:To study renal protective effect of the cryopreservation with oxygenated perfluorocar-bon preservation solutions.Methods:Male SD rats were randomly divided into (hypertonic citrate ade-nine)HCA and (perfluorocarbon emulsion)PFCE groups with twelve rats in each group.The kidney was infused in situ with the corresponding HCA solution and PFCE,respectively,then harvested and pre-served at 0~4℃in 20 ml of liquids for 24 hours.The levels of MDA,SOD activity and Bcl-2 protein as well as morphological changes were determined.Results:The renal MDA content was significantly lower in PFCE group than in HCA group (P<0.05 ),SOD activity and Bcl-2 expression were significantly higher in PFCE group than HCA group (P<0.05 ),pathological scores were lower in PFCE group than HCA group (P<0.05 ).Futhermore,PFCE reduced renal tubular epithelial cell injury compared with HCA solution.Conclusion:Perfluororcarbon emulsions can improve antioxidant and anti-apoptosis ability of kidney to reduce renal damage during preservation of donor kidney.
