目的:评价左旋聚乳酸/壳聚糖纳米纤维/纳米羟基磷灰石(poly (L-latic acid )/chitosan nano-fibers/nano-hydroxylapatite,PLLA/CSNF/nHA)复合仿生支架联合骨髓间充质干细胞(bone marrow mesenchymal stem cells, BMSCs)修复新西兰兔桡骨大段缺损的可行性。方法:二次相分离法制作PLLA/CSNF/nHA仿生支架,并与新西兰兔的3代BMSCs复合。制作新西兰兔桡骨1.5 cm段状缺损模型27只,实验组缺损处植入PLLA/CSNF/nHA/BM-SCs复合物、对照组植入PLLA/CSNF/nHA仿生支架、空白组不做处理,于术后第4、10、16周通过大体形态观察、放射学检查和组织学检查等方法评价缺损修复情况。结果:术后末次观察,实验组已完成缺损修复,材料完全降解;对照组部分修复,仍残留部分缺损;空白组缺损主要由纤维结缔组织填充。结论:PLLA/CSNF/nHA复合支架的仿生结构具有良好的生物相容性,联合BMSCs在生物体内可以加快新骨生成,完成新西兰兔大段骨缺损的修复。

목적:평개좌선취유산/각취당납미섬유/납미간기린회석(poly (L-latic acid )/chitosan nano-fibers/nano-hydroxylapatite,PLLA/CSNF/nHA)복합방생지가연합골수간충질간세포(bone marrow mesenchymal stem cells, BMSCs)수복신서란토뇨골대단결손적가행성。방법:이차상분리법제작PLLA/CSNF/nHA방생지가,병여신서란토적3대BMSCs복합。제작신서란토뇨골1.5 cm단상결손모형27지,실험조결손처식입PLLA/CSNF/nHA/BM-SCs복합물、대조조식입PLLA/CSNF/nHA방생지가、공백조불주처리,우술후제4、10、16주통과대체형태관찰、방사학검사화조직학검사등방법평개결손수복정황。결과:술후말차관찰,실험조이완성결손수복,재료완전강해;대조조부분수복,잉잔류부분결손;공백조결손주요유섬유결체조직전충。결론:PLLA/CSNF/nHA복합지가적방생결구구유량호적생물상용성,연합BMSCs재생물체내가이가쾌신골생성,완성신서란토대단골결손적수복。
Aim:To evaluate the feasibility of the biomimetic poly (L-latic acid )/chitosan nano-fi-bers/nano-hydroxylapatite (PLLA/CSNF/nHA)composite scaffolds combined with bone marrow mesen-chymal stem cells (BMSCs)to repair bone defect of New Zealand rabbits.Methods:PLLA/CSNF/nHA composite scaffolds were prepared through the dual phase separation technique,then co-cultured with BM-SCs.The large defects (1.5 cm)in radius of rabbits were prepared and experimented in three groups. PLLA/CSNF/nHA/BMSCs compound was planted into the defected position in the experimental group;PLLA/CSNF/nHA scaffolds in the control;and no implant in the blank.The curative effect was evalua-ted by general -observation,radiology examination and HE staining at the 4th,1 0th and 1 6th week.Re-sults:The defects were repaired perfectly with the scaffolds degraded in the experimental group;partially repaired and degraded in the control;and full of fibrillar connective tissues in the blank,by the last eval-uation.Conclusion:PLLA/CSNF/nHA biomimetic scaffolds composited with BMSCs of rabbits have a good role to accelerate the formation of new bone and repair the large bone defect.
