为研究葛根黄酮(RPF)对糖尿病模型小鼠糖脂代谢的影响及其可能的作用机制,采用四氧嘧啶(ALX,220mg/kg)腹腔注射建立糖尿病小鼠模型。RPF连续灌胃15d后摘眼球取血,分离血清,测定血糖、血清甘油三酯(TG)、总胆固醇(TC)、一氧化氮(NO)和一氧化氮合酶(NOS)水平;取肝、肾脏制成10%匀浆测定其NO和NOS水平;观察胰腺病理学形态变化。结果显示RPF能降低ALX诱导的糖尿病模型小鼠血糖、血清和肝脏中NO和NOS。其降糖机制可能与降低血清、肝脏的NO和NOS水平有关。

위연구갈근황동(RPF)대당뇨병모형소서당지대사적영향급기가능적작용궤제,채용사양밀정(ALX,220mg/kg)복강주사건립당뇨병소서모형。RPF련속관위15d후적안구취혈,분리혈청,측정혈당、혈청감유삼지(TG)、총담고순(TC)、일양화담(NO)화일양화담합매(NOS)수평;취간、신장제성10%균장측정기NO화NOS수평;관찰이선병이학형태변화。결과현시RPF능강저ALX유도적당뇨병모형소서혈당、혈청화간장중NO화NOS。기강당궤제가능여강저혈청、간장적NO화NOS수평유관。
To evaluate the effects of Radix Pueraria Flavonoids on blood glucose and lipid level of diabetic mice induced by alloxan and study the hypoglycemic activity of Radix Pueraria Flavonoids and the mechanism.Injecting alloxan(220 mg/kg) into its abdominal cavity to set animal model of diabetes.After 15d administration of RPF,blood glucose were determined and blood serum was drawn to determine TG,TC,NO and NOS.Liver and kidney were drawn to determine NO and NOS.Pancreatic pathology was studied with morphological method.Compared with diabetic model group,the results show that RPF could decrease the blood glucose level and lower NO,NOS in blood serum and liver.The mechanisms of hypoglycemic activity of RPF may be related to lowering NO and NOS in blood serum and liver.
