以1,10-菲罗啉、壳聚糖和四水合乙酸钴为原料,合成了水溶性壳聚糖钴(II)配合物(CHT-Co(II)),采用红外光谱对配体及配合物进行了表征。在pH 7.5的Tris-HCl缓冲溶液中,采用粘度法、紫外可见吸收光谱法和电化学方法(循环伏安法,微分脉冲伏安法)研究了配合物与DNA的相互作用。粘度法表明二者能发生部分嵌插作用;DNA的加入能引起配合物紫外光谱的变化也表明了二者之间的相互作用。电化学实验表明配合物在玻碳电极上的氧化还原为一可逆过程,当加入一定量的DNA时,配合物的氧化峰电流明显降低,式电位有所正移,验证了二者之间存在部分嵌插作用。

A cobalt complex with 1,10-phenanthroline and water-soluble chitosan(CHT-Co(Ⅱ)) as ligands was synthesized and characterized.In pH 7.5 Tris-HCl buffer solution,the interaction of the cobalt complex with DNA was investigated by viscosity UV-visible absorption spectroscopy,and electrochemical methods(cyclic voltammetry,differential pulse voltammetry).Viscosity experiments revealed that the cobalt complex bind to DNA via partial intercalation.UV-spectrum of the CHT-Co(Ⅱ) complex was changed when DNA were added,proving the interaction of the two species.The electrochemistry of the complex on a glassy carbon electrode showed a pair of reversible redox potential corresponding to the electron transfer of Co(Ⅱ)/Co(I) couple.When a certain amount of DNA was added,the peak current of the complex decreasedy,with a positive shift of peak potential,confirming the intercalative mode between the CHT-Co(Ⅱ) complex and DNA.
