目的:探讨小细胞肺癌c-myc癌基因的表达对Nrf2表达的影响,分析c-myc与Nrf2表达的相关性。方法:选用两株耐药性不同的肺小细胞癌肿瘤细胞株,小细胞肺癌药物敏感株H69及耐药株H69AR,Western-Blot方法比较Nrf2,c-myc在两株细胞间的表达差异;H69细胞通过Lipofectamine2000脂质体转染过表达c-myc,Western-Blot方法检测过表达c-myc前后Nrf2蛋白表达变化;H69AR细胞通过Lipofectamine2000脂质体转染c-myc siRNA, Western-Blot方法检测c-myc siRNA前后Nrf2的蛋白表达变化。结果:与H69细胞相比,H69AR细胞株中c-myc及Nrf2高表达;H69细胞c-myc转染后,Nrf2表达上调;H69AR细胞c-myc siRNA干扰后,Nrf2表达下调。结论:小细胞肺癌中Nrf2的表达与c-myc表达呈正相关。

목적:탐토소세포폐암c-myc암기인적표체대Nrf2표체적영향,분석c-myc여Nrf2표체적상관성。방법:선용량주내약성불동적폐소세포암종류세포주,소세포폐암약물민감주H69급내약주H69AR,Western-Blot방법비교Nrf2,c-myc재량주세포간적표체차이;H69세포통과Lipofectamine2000지질체전염과표체c-myc,Western-Blot방법검측과표체c-myc전후Nrf2단백표체변화;H69AR세포통과Lipofectamine2000지질체전염c-myc siRNA, Western-Blot방법검측c-myc siRNA전후Nrf2적단백표체변화。결과:여H69세포상비,H69AR세포주중c-myc급Nrf2고표체;H69세포c-myc전염후,Nrf2표체상조;H69AR세포c-myc siRNA간우후,Nrf2표체하조。결론:소세포폐암중Nrf2적표체여c-myc표체정정상관。
Objective:To determine the influence of oncogene c-myc over Nrf2 in small cell lung cancer, and to an-alyze the correlation between c-myc and Nrf2 expressions. Methods:Two lines of small-cell lung cancer cells were chosen in this study. One is drug sensitive H69 cell. The other is multidrug resistant H69AR cell. Western blot assays were uti-lized to compare the different expression levels of c-myc and Nrf2. Overexpression of c-myc by Lipofectamine 2000 trans-fection was performed in H69 cells, and Nrf2 protein expression was compared between the control and c-myc transfection groups. Down-regulation of c-myc by siRNA transfection was performed in H69AR cells, and Nrf2 protein expression was compared between the control and c-myc siRNA groups. Results: Compared with H69 cells, protein expression level of c-myc and Nrf2 was much higher in H69AR cell. In H69 tumor cells, Nrf2 was up-regulated after c-myc transfection. Mean-while, in H69AR tumor cells, Nrf2 was down-regulated after c-myc siRNA transfection. Conclusion: Nrf2 and c-myc is positively correlated in small cell lung cancer. To determine the influence of c-myc over Nrf2 in small cell lung cancer, and to analyse the correlation between c-myc and Nrf2 expressions.
