目的:探讨水甘油通道蛋白7与高脂膳食诱导的大鼠肥胖的关系。方法:雄性6周龄SD大鼠随机分为高脂饲料组24只,普通饲料组10只。8周后将高脂饲料组大鼠分为饮食诱导肥胖组(OP)和饮食诱导肥胖抵抗组(OR)大鼠,口服葡萄糖耐量实验检测各组大鼠的胰岛素敏感性。实验结束分离大鼠的肾周和睾周脂肪组织并准确称重,分离血清测定血糖血脂。实时荧光定量PCR技术检测大鼠脂肪组织AQP7的mRNA表达。结果:OP组大鼠体重(556.8±10.6)g、体脂(6.47±0.48)%、TC(1.43±0.14)mmoL/L、TG(0.78±0.13)mmoL/L、Insulin含量(66.09±8.06)μIU/mL和HOMA-IR (19.63±3.08)。显著高于OR组的(478.9±8.9)g、(4.53±0.43)%、(1.15±0.10)mmoL/L、(0.62±0.04)mmoL/L、(48.03±8.70)μIU/mL、(14.74±9.61)和对照组(470.5±8.6)g、(3.34±0.28)%、(1.17±0.15)mmoL/L、(0.51±0.08)mmoL/L、(29.57±6.94)μIU/mL、(8.08±0.81)。OP组AQP7的mRNA表达(3.80±0.38)与OR组(1.12±0.15)和对照组(1.00±0.00)相比也显著增加,OR组和C组相比差异无统计学意义。结论:脂肪组织中AQP7表达与高脂膳食诱导的肥胖密切相关。

Objective:To establish high fat diet induced obesity prone and obesity resistant rats and investigate the AQP7 expression in white adipose tissue in each group. Methods: six-week-old outbred male SD rats were randomly di-vided into two groups: the high-fat diet group (HF, n=24) and the chow food group (C, n=10). After 8 weeks, rats in HF group were designated as OP and OR rats. Insulin sensitivity was determined by oral glucose tolerance test. AQP7 mRNA expression was detected by Real time Polymerase Chain Reaction (RT-PCR). Results:Among OP , OR group and C group, body weights were (556.8±10.6), (478.9±8.9) and (470.5±8.6)g, (P<0.05 );body fat was (6.47±0.48), (4.53±0.43) and (3.34± 0.28)%, (P<0.05);Serum TC was (1.43±0.14), (1.15±0.10) and (1.17±0.15), (P<0.05);TG was (0.78±0.13), (0.62±0.04) and (0.51 ±0.08), (P<0.05); Insulin was (66.09 ±8.06), (48.03 ±8.70) and (29.57 ±6.94), (P<0.05); HOMA-IR was (19.63 ±3.08), (14.74±9.51) and (8.08±0.81), (P<0.05); AQP7 mRNA expression in white adipose tissue was (3.80±0.38), (1.12±0.15) and (1.00±0.00), (P<0.05). Conclusion: AQP7 mRNA expression in white adipose tissue is closely related to high-fat diet in-duced obesity.
