采用 SSR分子标记对迎春樱(Cerasus discoidea)4个居群共112个单株的遗传多样性进行了研究.结果显示:15对SSR引物共检测到54条等位基因,平均每位点等位基因3.6个,平均多态位点比率为86.67%.Nei′s基因多样性指数为0.3676~0.4990,Shannon信息指数为0.5883~0.8515.分子方差分析(AMOVA)表明,迎春樱78%的遗传变异来自居群内,22%的遗传变异来自于居群间.居群间基因流(Nm)为0.6944,遗传分化系数(Fst)为0.264,表明迎春樱居群间的遗传分化水平较高,基因交流受阻.聚类分析结果显示,4个居群可聚为2支.对迎春樱居群中等遗传多样性和居群间较高遗传分化形成的可能原因进行了分析,并提出了迎春樱的保护利用策略.

Genetic diversity of 1 1 2 individuals from 4 Cerasus discoidea populations was investigated u-sing 15 SSR markers.A total of 54 alleles were amplified,and the mean number of alleles per locus was 3.6,the average of polymorphic locus percentage was 86.67%.The Nei's diversity index was 0.367 6~0.499 0 and Shannon's diversity was 0.588 3~0.851 5.The AMOVA analysis showed that the majority of genetic variation occurred within populations(78% ),and 22% occurred among the populations.The coeffi-cient of genetic differentiation (Fst)and gene flow (Nm)were 0.264 and 0.694 4,which indicated that the genetic differentiation was relatively evident,with fewness gene flow in Cerasus discoidea populations.The 4 populations were divided into 2 clades according to genetic distance.The possible formation of the middle level genetic diversity and the high genetic differentiation were discussed,and further strategies and sug-gestions for utilization and conservation of these resources were proposed.
