为研究牛四倍体胚胎制备的条件及发育程度,以牛卵母细胞为试验材料进行体外受精(in vitro fertiliza-tion,IVF),受精后统计处于2-细胞期的胚胎数量,选取3个组合进行2-细胞期胚胎的融合,并统计融合率和融合后四倍体胚胎的存活率、卵裂率和囊胚率.结果表明:受精卵培养至14 h时卵裂率最高,达到46.28%,因而确定为四倍体胚胎制备的开始时间.100 V/mm,20μs,1次脉冲组的融合率(67.92±9.76)%、四倍体胚胎的存活率(93.71±5.31)%、卵裂率(70.37±7.07)%和囊胚率(19.44±3.62)%都显著高于其他2个组.因此,牛体外受精卵在mCR1aa培养液中培养14 h后用100 V/mm,20μs,1次脉冲进行四倍体的制备效果最佳.

The aim of present study was to investigate best conditions for generating tetroploid embryos ac-cording to the cleavage rate and subsequent embryonic development during bovine invitro fertilization (IVF).All the 2-cell embryos were counted and fused to generate tetroploid embryos after IVF.The fusion rate,survival rate, cleavage rate and blastocyst development of tetroploid embryos were evaluated.The results showed that the highest cleavage rate (46.28%)of zygotes appeared at 14 hours after in vitro culture.The fusion rate (67.92±9.76)%, survival rate (93.71± 5.31)%,cleavage rate (70.37±7.07)% and blastocyst rate (19.44±3.62)% of tetroploid embryos in the group with a single direct pulse of 100 V/mm for 20μs was significantly higher than other groups. Therefore,it was best to generate tatroploid embryos from bovine blastomeres that were cultured for 1 4 h and fused with a single direct pulse of 100 V/mm for 20μs.
