目的:通过对膝骨性关节炎( KOA)模型大鼠的实验,观察复方中药骨炎康颗粒对膝骨性关节炎的作用机制。方法选用SD大鼠,随机分为正常对照组(A组)、模型组(B组)、骨炎康颗粒治疗组(C组)、西药对照组(D组)。通过左膝关节腔内注入木瓜蛋白酶,造成骨性关节炎病变模型。木瓜蛋白酶注射结束后2周,开始对C、D组分别予以药物骨炎康颗粒、布洛芬灌胃,8周后处死全部大鼠,取A组双侧膝关节及B、C、D组造模侧膝关节制作病理标本,分别行免疫组化( TGF-β1、IL-lβ)染色,观察血清II型胶原、关节软骨及白介素l(1L-lβ)、转移生长因子(TGF-β1)的改变。结果(1)血清II型胶原结果:骨炎康颗粒能提高血清II型胶原含量;(2)TGF-β1、IL-lβ改变结果:各治疗组均能提高TGF-β1含量,并降低1L-lβ,其中以骨痹颗粒组效果最佳,布洛芬相对较弱。结论骨炎康颗粒可增进软骨基质的合成,抑制软骨基质的分解,促进受损关节软骨的修复。骨炎康颗粒的疗效机理可能在于通过对机体的细胞因子调节,提高血清II型胶原、关节软骨的TGF-β1含量,降低IL-lβ含量,从而直接或间接促进了损伤关节软骨的修复。

목적:통과대슬골성관절염( KOA)모형대서적실험,관찰복방중약골염강과립대슬골성관절염적작용궤제。방법선용SD대서,수궤분위정상대조조(A조)、모형조(B조)、골염강과립치료조(C조)、서약대조조(D조)。통과좌슬관절강내주입목과단백매,조성골성관절염병변모형。목과단백매주사결속후2주,개시대C、D조분별여이약물골염강과립、포락분관위,8주후처사전부대서,취A조쌍측슬관절급B、C、D조조모측슬관절제작병리표본,분별행면역조화( TGF-β1、IL-lβ)염색,관찰혈청II형효원、관절연골급백개소l(1L-lβ)、전이생장인자(TGF-β1)적개변。결과(1)혈청II형효원결과:골염강과립능제고혈청II형효원함량;(2)TGF-β1、IL-lβ개변결과:각치료조균능제고TGF-β1함량,병강저1L-lβ,기중이골비과립조효과최가,포락분상대교약。결론골염강과립가증진연골기질적합성,억제연골기질적분해,촉진수손관절연골적수복。골염강과립적료효궤리가능재우통과대궤체적세포인자조절,제고혈청II형효원、관절연골적TGF-β1함량,강저IL-lβ함량,종이직접혹간접촉진료손상관절연골적수복。
Objective To investigate the mechanism of Guyankang granules, a compound traditional Chinese medicine, on knee osteoarthritis (KOA) rat model.Methods SD rats were selected and randomly divided into normal control group (Group A), model group ( Group B ) , Guyankang granules treated group ( Group C ) , and western medicine group ( Group D ) .The rat osteoarthritis model was established using the injection of papain into the left knee joint cavity.Two weeks after the injection of papain, rats in Group C and Group D were given a gavage of Guyankang granules and ibuprofen, respectively.Eight weeks later, all the rats were executed.The bilateral knees in Group A and model lateral knee in Group B, Group C, and Group D were collected and made into pathological specimen, respectively.The immunohistochemical staining of transforming growth factorβ1 (TGF-β1) and interleukin 1β( IL-lβ) was performed.The changes of serum type II collagen, the articular cartilage, IL-lβ, and TGF-β1 were observed.Results Guyankang granules could improve the content of serum type II collagen.The results of the changes of TGF-β1and IL-lβrevealed that the content of TGF-β1 in each treatment group increased significantly, while the content of IL-lβreduced. The effect in Guyankang Group was the best, while the effect in ibuprofen group was relatively weak.Conclusion Guyankang granules can promote the synthesis of the cartilage matrix, inhibit the decomposition of the cartilage matrix, and promote the repair of the damaged articular cartilage.The possible mechanism of Guyankang granules may be the regulation of cytokines, thus improving the content of serum type II collagen and the content of TGF-β1 in the articular cartilage, reducing the content of IL-lβ, then promoting the repair of the injured articular cartilage directly or indirectly.
