目的初步探讨趋化因子基质细胞衍生因子-1α( SDF-1α)及其受体CXCR4在有血液系统损害的系统性红斑狼疮( SLE)患者外周血的表达水平及意义。方法用流式细胞术分别检测20名健康对照者、15例SLE非血液系统损害患者,25例SLE血液系统损害患者外周血中CD3+CD4+T淋巴细胞、CD3+CD8+T淋巴细胞表面CXCR4的表达百分率,采用双抗体夹心ABC-ELISA法检测以上各组血清SDF-1α浓度水平。结果健康对照者外周血中CD3+CD4+T淋巴细胞、CD3+CD8+T淋巴细胞表面CXCR4表达百分率及血清SDF-1α浓度水平均明显低于SLE非血液系统损害患者和SLE血液系统损害患者( P<0.05),而SLE非血液系统损害患者外周血中CD3+CD4+T淋巴细胞、CD3+CD8+T淋巴细胞表面CXCR4表达百分率及血清SDF-1α浓度水平也均明显低于SLE血液系统损害患者( P=0.000)。 SLE血白细胞减少患者、血小板减少患者、贫血患者的外周血中SDF-1α、CXCR4表达水平也较SLE非血白细胞减少患者、血小板减少患者、贫血患者高。结论 SDF-1α、CXCR4可能在SLE血液系统损害的发病过程中起重要作用。

목적초보탐토추화인자기질세포연생인자-1α( SDF-1α)급기수체CXCR4재유혈액계통손해적계통성홍반랑창( SLE)환자외주혈적표체수평급의의。방법용류식세포술분별검측20명건강대조자、15례SLE비혈액계통손해환자,25례SLE혈액계통손해환자외주혈중CD3+CD4+T림파세포、CD3+CD8+T림파세포표면CXCR4적표체백분솔,채용쌍항체협심ABC-ELISA법검측이상각조혈청SDF-1α농도수평。결과건강대조자외주혈중CD3+CD4+T림파세포、CD3+CD8+T림파세포표면CXCR4표체백분솔급혈청SDF-1α농도수평균명현저우SLE비혈액계통손해환자화SLE혈액계통손해환자( P<0.05),이SLE비혈액계통손해환자외주혈중CD3+CD4+T림파세포、CD3+CD8+T림파세포표면CXCR4표체백분솔급혈청SDF-1α농도수평야균명현저우SLE혈액계통손해환자( P=0.000)。 SLE혈백세포감소환자、혈소판감소환자、빈혈환자적외주혈중SDF-1α、CXCR4표체수평야교SLE비혈백세포감소환자、혈소판감소환자、빈혈환자고。결론 SDF-1α、CXCR4가능재SLE혈액계통손해적발병과정중기중요작용。
Objective To explore the expression of SDF-1αand CXCR4 in the peripheral blood of hematological damage in systemic lupus erythematosus ( SLE ) .Methods Forty hospitalized SLE patients were recruited and twenty healthy volunteers were enrolled as normal controls .The expression of SDF-1αand CXCR4 in control group(20 cases),hematological damage group (25 cases) and no damage group (15 cases) were detected by ELISA and flow cytometry .Results Compared with the normal control group , the damage and no damage groups demonstrated the significantly higher levels of SDF-1α,CD3 +CD4 +CXCR4 +and CD3 +CD8 +CXCR4 +( P<0.05 ) . Furthermore,the no damage group showed significantly lower levels of SDF-1α, CD3 +CD4 +CXCR4+ and CD3 +CD8 +CXCR4 +than damage group(P=0.000).In addition,the expressions of SDF-1αand CXCR4 in the anemia group,low white blood cell group and thrombocytopenia group were significantly higher than no damage group . Conclusion SDF-1α/CXCR4 may play an important role in the pathogenesis of hematological damage in SLE .
