Wuyu decoction%ovarian granulose cell%P38 MAPK
目的:运用血清药理学方法观察乌萸汤含药血清对大鼠卵巢颗粒细胞P38丝裂原激活的蛋白激酶(MAPK)信号转导途径的影响。方法:25只SD大鼠随机分为正常对照组、西药对照组、乌萸汤低、中、高剂量组,给药4 d后采血并分离血清。然后在大鼠卵巢颗粒细胞培养体系中加入各组血清,培养48 h后,用Western blot测定颗粒细胞P38 MAPK磷酸化蛋白的相对表达。结果:与空白对照组比较,各给药组颗粒细胞P38 MAPK磷酸化蛋白相对表达量明显增多(P<0.01),乌萸汤中剂量组效果优于西药组(P<0.01)。结论:乌萸汤含药血清通过提高P38 MAPK蛋白磷酸化水平从而促进卵巢颗粒细胞增殖,表明乌萸汤是通过调控P38 MAPK信号转导途径以促进卵泡发育成熟及排出。

목적:운용혈청약이학방법관찰오유탕함약혈청대대서란소과립세포P38사렬원격활적단백격매(MAPK)신호전도도경적영향。방법:25지SD대서수궤분위정상대조조、서약대조조、오유탕저、중、고제량조,급약4 d후채혈병분리혈청。연후재대서란소과립세포배양체계중가입각조혈청,배양48 h후,용Western blot측정과립세포P38 MAPK린산화단백적상대표체。결과:여공백대조조비교,각급약조과립세포P38 MAPK린산화단백상대표체량명현증다(P<0.01),오유탕중제량조효과우우서약조(P<0.01)。결론:오유탕함약혈청통과제고P38 MAPK단백린산화수평종이촉진란소과립세포증식,표명오유탕시통과조공P38 MAPK신호전도도경이촉진란포발육성숙급배출。
Objective:To research the effects of Wuyu decoction drug-containing serum on expression of P38 MAPK of ovarian granulosa cells (GC) of rats by serum pharmacological method. Methods:25 SD rats were randomly divided into normal con-trol group,western medicine group,low,middle and high dosage of Wuyu decoction group. Blood was taken from femoral artery of rats and serums were separated 4 d after administration. Serum in each group was added into ovarian granulose cell medium system of rats,then 48 h later relative expression of P38 MAPK phosphorylated protein was measured by Western blot. Results:Compared with blank control group,relative expression of P38 MAPK phosphorylated protein was significantly increased in treatment groups (P<0.01). Effect of middle dosage group of Wuyu decoction was better than that of western medicine group (P<0.01). Conclusion:Wuyu decoction drug-containing serum could promote the proliferation of ovarian granulosa cells through enhancing P38 MAPK protein phosphorylation. It is indicated that Wuyu decoction cou ld promote development,maturation and ovulation of ovarian follicle through adjusting P38 MAPK signal transduction pathway.
