背景:研究表明,间充质干细胞是重要的免疫调节细胞,在自身免疫病的治疗中有潜在的应用前景。<br> 目的:探讨间充质干细胞移植在治疗多发性硬化症等自身免疫性疾病中的机制。<br> 方法:用小鼠制备间充质干细胞,并通过静脉将间充质干细胞输入同基因、异基因小鼠体内,观察间充质干细胞对小鼠中枢免疫器官(胸腺)、外周免疫器官(脾脏)中调节T细胞(Treg)的百分比、Foxp3及白细胞介素10和转化生长因子β1的mRNA的表达。<br> 结果与结论:同基因及异基因间充质干细胞移植均能促进小鼠脾脏、胸腺及淋巴结中CD4+CD25+Foxp3+T细胞表达水平显著增高,促进小鼠脾脏、胸腺及淋巴结中Foxp3、白细胞介素10和转化生长因子β1的mRNA表达水平显著增高。提示间充质干细胞移植具有治疗自身免疫性疾病的作用,该作用可能是由CD4+CD25+Foxp3+T细胞、白细胞介素10、转化生长因子β1及Foxp3的高表达所介导。

BACKGROUND:Mesenchymal stem cells are found to have the immunoregulatory activities and a potential application prospect in the treatment of autoimmune diseases. <br> OBJECTIVE:To explore the mechanism of transplanting mesenchymal stems cells on the treatment of multiple sclerosis. <br> METHODS:The mouse mesenchymal stems cells were prepared, and injected into the al ogenic and syngenic normal mice, to detect the frequency of CD4+CD25+Foxp3+T cells in the spleen, thymus, and lymph nodes by flow cytometry, and to detect the Foxp3, transforming growth factor-β1, and interleukin-10 mRNA in the spleen, thymus, and lymph nodes by reverse transcription-PCR. <br> RESULTS AND CONCLUSION:Transplantation of mesenchymal stem cells on normal mice led to a significant up-regulation of CD4+CD25+Foxp3+T cells, Foxp3, transforming growth factor-β1, and interleukin-10 mRNA in the spleen, thymus, and lymph nodes both in the al ogenic and syngenic transplant groups. Transplantation of mesenchymal stem cells may be an available method in the treatment of autoimmune diseases, and CD4+CD25+Foxp3+T cell, Foxp3, transforming growth factor-β1, and interleukin-10 may be involved in this process.
