DHA-PC%mice%sleep
目的:研究二十二碳六烯酸-磷脂( DHA-PC )对小鼠睡眠的改善作用。方法小鼠随机分为正常对照组、溶剂对照组、DHA鱼油+卵磷脂联合用药(DHA+Lecithin)(60+200)mg/kg(阳性对照)、DHA-PC 50、100、200 mg/kg组,每组10只。正常对照组小鼠自由饮食饮水,溶剂对照组小鼠按0.2 ml/10 g的体积灌胃给予生理盐水, DHA+Lecithin(60+200)mg/kg组和DHA-PC 50、100、200 mg/kg组小鼠均按0.2 ml/10 g的体积灌胃给药,1次/d,连续30 d。通过测定小鼠直接睡眠时间、戊巴比妥钠诱导睡眠时间、阈下剂量睡眠发生率及巴比妥钠诱导睡眠潜伏时间,观察DHA-PC对小鼠睡眠的作用。结果3种剂量的DHA-PC对小鼠体质量均无影响。直接睡眠实验结果表明,3种剂量的DHA-PC对小鼠直接睡眠无影响。戊巴比妥钠诱导睡眠实验结果表明, DHA-PC 50、100、200 mg/kg组小鼠戊巴比妥钠诱导睡眠时间分别为(56.2±13.7),(57.9±25.4)及(64.1±18.4)min,明显高于溶剂对照组(32.9±10.8) min ( P <0.05);DHA +Lecithin (60+200) mg/kg 组小鼠的戊巴比妥钠睡眠时间为(38.6±11.7)min,高于溶剂对照组小鼠(32.9±10.8)min,但无明显差异;DHA-PC 200mg/kg组明显高于DHA+Lecithin (60+200)mg/kg组(P<0.05)。协同戊巴比妥钠阈下剂量催眠实验结果表明,DHA-PC 200 mg/kg组小鼠协同戊巴比妥钠阈下剂量催眠入睡率(70%)明显高于溶剂对照组(10%)(P<0.05);DHA+Lecithin (60+200) mg/kg组小鼠协同戊巴比妥钠阈下剂量催眠入睡率(60%)明显高于溶剂对照组(10%)( P<0.05)。巴比妥钠诱导睡眠潜伏期实验结果表明,DHA-PC 200 mg/kg和DHA+Lecithin (60+200) mg/kg组小鼠巴比妥钠诱导睡眠潜伏时间分别为(22.9±4.1)和(19.5±2.7)min,明显低于溶剂对照组(31.3±6.9)min(P<0.01),且DHA+Leci-thin(60+200)mg/kg组小鼠巴比妥钠诱导睡眠潜伏时间为(19.5±2.7)min,明显低于DHA-PC 50和100 mg/kg组,二者分别为(27.3±2.4)和(26.9±4.2)min。结论 DHA-PC具有改善小鼠睡眠的作用。

목적:연구이십이탄륙희산-린지( DHA-PC )대소서수면적개선작용。방법소서수궤분위정상대조조、용제대조조、DHA어유+란린지연합용약(DHA+Lecithin)(60+200)mg/kg(양성대조)、DHA-PC 50、100、200 mg/kg조,매조10지。정상대조조소서자유음식음수,용제대조조소서안0.2 ml/10 g적체적관위급여생리염수, DHA+Lecithin(60+200)mg/kg조화DHA-PC 50、100、200 mg/kg조소서균안0.2 ml/10 g적체적관위급약,1차/d,련속30 d。통과측정소서직접수면시간、무파비타납유도수면시간、역하제량수면발생솔급파비타납유도수면잠복시간,관찰DHA-PC대소서수면적작용。결과3충제량적DHA-PC대소서체질량균무영향。직접수면실험결과표명,3충제량적DHA-PC대소서직접수면무영향。무파비타납유도수면실험결과표명, DHA-PC 50、100、200 mg/kg조소서무파비타납유도수면시간분별위(56.2±13.7),(57.9±25.4)급(64.1±18.4)min,명현고우용제대조조(32.9±10.8) min ( P <0.05);DHA +Lecithin (60+200) mg/kg 조소서적무파비타납수면시간위(38.6±11.7)min,고우용제대조조소서(32.9±10.8)min,단무명현차이;DHA-PC 200mg/kg조명현고우DHA+Lecithin (60+200)mg/kg조(P<0.05)。협동무파비타납역하제량최면실험결과표명,DHA-PC 200 mg/kg조소서협동무파비타납역하제량최면입수솔(70%)명현고우용제대조조(10%)(P<0.05);DHA+Lecithin (60+200) mg/kg조소서협동무파비타납역하제량최면입수솔(60%)명현고우용제대조조(10%)( P<0.05)。파비타납유도수면잠복기실험결과표명,DHA-PC 200 mg/kg화DHA+Lecithin (60+200) mg/kg조소서파비타납유도수면잠복시간분별위(22.9±4.1)화(19.5±2.7)min,명현저우용제대조조(31.3±6.9)min(P<0.01),차DHA+Leci-thin(60+200)mg/kg조소서파비타납유도수면잠복시간위(19.5±2.7)min,명현저우DHA-PC 50화100 mg/kg조,이자분별위(27.3±2.4)화(26.9±4.2)min。결론 DHA-PC구유개선소서수면적작용。
Objective To study the sleep improvement function of DHA-PC.Methods The mice were randomly divid-ed into control, vehicle, DHA+Lecithin (60+200 mg/kg) and DHA-PC(50,100,200 mg/kg) groups.Ten mice were enrolled in each group .The mice of control were administered with normal food , the vehicle group was orally given normal saline at the dosage of 0.2 ml/10 g, while both DHA-PC and DHA+Lecithin were orally given corresponding drugs at the dosage of 0.2 ml/10 g.All the groups were treated for 30 days except control group .The direct sleep-inducing test, the test of lengthening sleep time induced by pentobarbital sodium , the test of pentobarbital sodium subthreshold-hypnosis and the test of barbital sodium sleep latency were conducted to observe the inductive effect of DHA -PC.Results Neither the effect on mice body mass nor directly-induced sleep was observed .DHA-PC (50,100, and 200 mg/kg) could prolong sleep time to (56.2 ±13.7),(57.9 ±25.4) and(64.1 ±18.4) min, respectively,compared to vehicle(32.9 ±10.8)min (P<0.05).DHA+Lecithin could not prolong sleep time (38.6 ±11.7)min compared to (32.9 ±10.8)min of vehicle.There was significant difference compared with DHA-PC at the dosage of 200 mg/kg (64.1 ±18.4)min (P<0.05).DHA-PC (200 mg/kg) enhanced pentobarbital sodium subthreshold-hypnosis (70%) compared to vehicle (10%) (P<0.05),so did DHA+Lecithin (60%) compared to vehicle (10%) (P<0.05).Both DHA-PC (200 mg/kg)[(22.9 ±4.1)min ] and DHA+Lecithin [(19.5 ±2.7) min ]could shorten sleep latency compared to vehicle (31.3 ±6.9) min(P<0.01), and the sleep latency of DHA +Lecithin (19.5 ±2.7) min was shorter than that of DHA-PC(50,100 mg/kg).Conclusion DHA-PC has some effect some sleep improvement in mice .
