目的:探讨AICDA基因多态性与新生儿脐血总IgE水平的相关性。方法选取无过敏史足月产妇97例,有过敏史足月产妇70例,提取脐血基因组DNA,采用聚合酶链反应-高温连接酶反应(PCR-LDR)检测AICDA基因rs2518144、rs2028373、rs11046349位点的基因型。同时,采用电化学发光免疫分析法(ECLIA)检测新生儿脐血总IgE水平。结果新生儿脐血AICDA基因rs2518144位点3组基因型(AA/AG/GG)及rs11046349位点3组基因型(TT/TG/GG)分布频率及等位基因频率在无过敏史组和有过敏史组产妇间差异无统计学意义(P均>0.05)。rs2028373位点3组基因型(AA/AG/GG)及等位基因分布频率与对照组比较有明显差异(P值分别为0.020和0.006)。各位点基因型与脐血IgE水平无相关性(P均>0.05)。结论新生儿脐血rs2028373基因多态性与母亲过敏史有一定相关性;但rs2518144、rs2028373、rs11046349位点基因型与脐血IgE水平无相关性。

목적:탐토AICDA기인다태성여신생인제혈총IgE수평적상관성。방법선취무과민사족월산부97례,유과민사족월산부70례,제취제혈기인조DNA,채용취합매련반응-고온련접매반응(PCR-LDR)검측AICDA기인rs2518144、rs2028373、rs11046349위점적기인형。동시,채용전화학발광면역분석법(ECLIA)검측신생인제혈총IgE수평。결과신생인제혈AICDA기인rs2518144위점3조기인형(AA/AG/GG)급rs11046349위점3조기인형(TT/TG/GG)분포빈솔급등위기인빈솔재무과민사조화유과민사조산부간차이무통계학의의(P균>0.05)。rs2028373위점3조기인형(AA/AG/GG)급등위기인분포빈솔여대조조비교유명현차이(P치분별위0.020화0.006)。각위점기인형여제혈IgE수평무상관성(P균>0.05)。결론신생인제혈rs2028373기인다태성여모친과민사유일정상관성;단rs2518144、rs2028373、rs11046349위점기인형여제혈IgE수평무상관성。
Objective To investigate the correlation between total immunoglobulin E(IgE)levels in cord blood and activation-induced cytidine deaminase(AICDA)gene polymorphism.Methods Cord blood samples of 70 infants whose mothers had allergic diseases and 97 infants whose mothers had no history of allergic diseases were collected.Genomic DNA were extracted from cord blood samples,and polymerase chain reaction-ligase detection reaction (PCR-LDR)was used to detect rs251 81 44,rs2028373,rs1 1 046349 sites in AICDA gene.Meantime,total IgE levels in cord blood were detected by electro-chemiluminescence immunoassay (ECLIA ).Results There was no significant difference in distribution and allelic distribution frequencies of 3 rs251 81 44 genotypes (AA/AG/GG)and 3 rs1 1 046349 genotypes (TT/TG/GG)between allergic disease group and non-allergic disease group(P>0.05).However,significant difference in genotype distribution and allelic distribution frequencies of 3 rs2028373 genotypes (AA/AG/GG)between the 2 groups was detected(P=0.020 and 0.006 ).The genotyps of rs251 81 44,rs2028373 and rs1 1 046349 sites were not related with total IgE levels(P>0.05).Conclusions rs2028373 gene polymorphism is significantly correlated with allergic diseases,but the genotypes of rs251 81 44,rs2028373 and rs1 1 046349 sites are not correlated with total IgE levels.
