利用ISSR分子标记技术,对采自景东、易门、昆明的66个塔拉样品进行遗传多样性分析。结果表明:(1)从104对引物中筛选出来11对多态性明显、条带清晰、反应稳定的引物,11对引物共扩增出74条带,每个引物平均扩增出6.37条带,其中多态性条带数为5.55条,多态性条带比例为82.65%;(2)经软件分析得知,等位基因数值(Na)为1.8784±0.3291,有效位点数值(Ne)为1.4263±0.3420,基因多样性指数值(H)为0.2576±0.1773,香农指数值(I)为0.3947±0.2416,多态位点百分数为87.84%;(3)试验样品的遗传相似系数范围为0.5135~0.9865,具有较丰富的遗传多样性,聚类分析可分为6类。

An experiment was conducted to study the genetic diversity of 66 Tara samples from Jingdong , Yimen and Kunming by ISSR markers.11 polymorphic primers with obvious, clear and stable reaction are from 104 ISSR primers.There are 74 bands amplified with 11 primers.Each primer amplifies 6.37 bands on average.They contain 5.55 polymorphic bands and their polymorphism percentage is 82.65%.The numerical allele is Na=1.878 4±0.329 1, the effective site numerical is Ne=1.426 3±0.342 0, gene diversity index is H=0.257 6±0.177 3, Shannon index numerical is I=0.394 7±0.241 6, and the percentage of polymorphic loci is 87.84%.The range of the genetic similarity coefficient is 0.513 5-0.986 5 with abundant genetic diversity.66 materials can be divided into six categories by clustering methods.
