目的:检测PTEN蛋白和PCNA在原发性小肠腺癌组织、癌旁组织及正常小肠组织中的表达,探讨其与原发性小肠腺癌的关系,为原发性小肠腺癌预后判断提供依据。方法采用SP免疫组化法对32例小肠腺癌及其相应癌旁组织、8例正常小肠组织进行PTEN蛋白及PCNA的定位观察。结果原发性小肠腺癌组织中PTEN蛋白阳性率为71.88%(23/32),相应癌旁组织及正常小肠组织中PTEN蛋白未阳性率分别为15.63%(5/32)、2.50%(1/40),2组比较差异有统计学意义( P <0.05);原发性小肠腺癌组织中PTEN蛋白的表达与其分化程度及有无淋巴结转移相关( P <0.05)。原发性小肠腺癌组织中PCNA阳性率为65.63%(21/32),相应癌旁组织6.25%(2/32)及正常组织PCNA阳性率为2.50%(1/40),二者比较差异有统计学意义( P <0.05);原发性小肠腺癌组织中PCNA的表达与肿瘤部位、浸润深度、分化程度相关( P <0.05)。结论小肠腺癌中PTEN蛋白和PCNA的高表达可作为判断预后的有效指标,两者联合检测可早期诊断小肠腺癌并判断预后。

Objective To detect PTEN protein and proliferatingcell nuclear antigen (PCNA) in primary small bowel adencareinoma(SBAC) tissue,adjacent nontumorons tissue,and normal small bowel tissue,to investigate the relationship between the expression of tumor suppressor gene PTEN protein and PCNA and the biologcal behaviors of primary small bowel adencareinoma.Methods Expression of PTEN protein and PCNA location were examined by streptavidin-peroxidase immunohistochemistry method in 32 SBAC,the corresponding adjacent nontumorons tissue and 8 normal small bowel tissues. Results Expression of PTEN protein was detected in 71.88% of the primary SBAC tissue,while expression in adjacent nontumorous tissue and normal small bowel tissue was detected in 15.63%and 12.50%( P <0.05).The positive expression of PTEN was significantly related to the cell differentiation ,diversion of ganglia lymphatica in primary SBAC tissue ( P <0.05).Expression of PCNA was detected in 65.63%of the primary SBAC tissue.6.25%of the adjacent nontumor tissue and 2.50%normal small bowel tissue ( P <0.05);The positive expression 0f PCNA was a significantly related to the tumor location,tumor infiltration depth and cell differentiation in primary SBAC tissue ( P <0.05).Conclusion Expression of tumor suppressor gene PTEN and PCNA could be helpful in predicting the biological behavior and prognosis of primary SBAC.
