目的:观察弥漫性脑损伤大鼠脑水肿及损伤区皮层MMP-9和咬合蛋白表达的变化。方法成年健康SD雄性大鼠160只,按随机数字表法分为对照组( n =60)和模型组( n =100)。应用Marmarou’s法建立弥漫性脑损伤大鼠模型。2组分别于术后3、12、24、72、144 h行干湿重法检测2组大鼠脑组织含水量变化,行免疫组化和Western blot检测损伤区周围皮层MMP-9和咬合蛋白表达水平。对各结果进行相关性分析。结果与对照组比较,模型组各时间点大鼠脑组织含水量明显增加,脑组织损伤区周围皮层MMP-9表达明显增高( P <0.05),咬合蛋白表达明显降低( P <0.05)。脑组织含水量变化与MMP-9表达变化呈正相关( P <0.05),脑组织含水量变化与咬合蛋白表达变化呈负相关( P <0.05),MMP-9蛋白表达变化与咬合蛋白表达变化呈负相关( P <0.05)。结论弥漫性脑损伤发生后损伤区皮层MMP-9蛋白表达明显增加,咬合蛋白表达明显降低,脑水肿形成。 MMP-9表达的增加,导致了咬合蛋白破坏,脑水肿形成。

목적:관찰미만성뇌손상대서뇌수종급손상구피층MMP-9화교합단백표체적변화。방법성년건강SD웅성대서160지,안수궤수자표법분위대조조( n =60)화모형조( n =100)。응용Marmarou’s법건립미만성뇌손상대서모형。2조분별우술후3、12、24、72、144 h행간습중법검측2조대서뇌조직함수량변화,행면역조화화Western blot검측손상구주위피층MMP-9화교합단백표체수평。대각결과진행상관성분석。결과여대조조비교,모형조각시간점대서뇌조직함수량명현증가,뇌조직손상구주위피층MMP-9표체명현증고( P <0.05),교합단백표체명현강저( P <0.05)。뇌조직함수량변화여MMP-9표체변화정정상관( P <0.05),뇌조직함수량변화여교합단백표체변화정부상관( P <0.05),MMP-9단백표체변화여교합단백표체변화정부상관( P <0.05)。결론미만성뇌손상발생후손상구피층MMP-9단백표체명현증가,교합단백표체명현강저,뇌수종형성。 MMP-9표체적증가,도치료교합단백파배,뇌수종형성。
Objective To observe the changes of expression of MMP-9 and occludin in brain cortex of rats with diffuse brain injury and the changes of brain edema around injury region .Methods The 160 Sprague-Dawley adult male rats were randomly divided into control group ( n =60) and model group ( n =100).The animal models with diffuse brain injury were established in rats by means of Marmarou ’s method.At 3h,12h,24h,72h and 144h after injury,brain water content was detected by dry wet weight measurement method in both groups ,and the expression levels of MMP-9 and occludin around the injury area were measured by immunohistochemistry and Western Blot .Finally the correlation analysis was performed about these results.Results As compared with that in control group , the brain water content in model group was significantly increased in different time points , and the expression levels of MMP-9 around injury area were obviously increased ( P <0.05), however, the content of occludin was significantly decreased .The changes of brain water content were positively correlated to those of MMP-9 ( P <0.05).However the changes of brain water content were negatively correlated to those of occludin expression levels ( P <0.05),so were between MMP-9 expression and occludin expression .Conclusion After the diffuse traumatic brain injury , the expression levels of MMP-9 in brain cortex around injury area are obviously increased , however ,the expression levels of occluding are significantly decreased , and brain edema appears .The increase of MMP-9 expression levels results in the decrease of occludin expression levels ,as a result,brain edema occurs .
