目的:探讨硫氧还原蛋白过氧化物酶3(PRDX3)在前列腺癌早期诊断及恶性判断中的作用。方法通过蛋白组学对比前列腺癌组织和癌旁组织中PRDX3基因结合蛋白差异表达程度,免疫组化验证,结合PRDX3免疫组化评分和前列腺癌患者的临床病理参数进行分析。结果通过蛋白组学结果表明,与癌旁组织相比,PRDX3在肿瘤组织中表达是上调的,上调达1.68倍( P=0.015)。通过免疫组化染色,我们发现PRDX3在前列腺癌组织中的表达明显强于癌旁组织( P=0.035)。 PRDX3的表达与患者年龄( P=0.028)有关;与前列腺癌患者血清PSA水平( P=0.641)和肿瘤转移( P=0.21)无明显相关;而与Gleason评分( P=0.040)、临床分期( P=0.024)和病理分期( P=0.041)有关。结论 PRDX3的表达与肿瘤的早期诊断和恶性程度相关,可结合ROS水平对前列腺癌疑似患者进行早期诊断及初步判断肿瘤恶性程度。

Objective To explore the role of Peroxiredoxin-3( PRDX3) in early diagnosis of prostate cancer ( PCa) and malignant judgment .Methods Analyzing the differentially expressed degree of PRDX 3 protein in PCa and adjacent tissues by 3D-DIGE technology,and then immunohistochemistry (IHC) validation,the combination of PRDX3 IHC score and clinicopathological parameters of PCa patients were analyzed .Results We found that PRDX3 expression was up-regulated in PCa tissues by proteomics analyzing ( Log Rank =1.68 , P =0.015 ) compared to adjacent tissues.The expression of PRDX3 in the adjacent tissues significantly stronger than PCa tissues ( P =0.035 ) .It was correlation with age ( P =0.028 ) , Gleason score ( P =0.040 ) , clinical stages ( P =0.024 ) and pathological stages ( p=0.041 ) , but not serum PSA levels ( P=0.0641 ) and tumor metastasis .Conclusion The expression of PRDX3 is closely related with the malignancy of PCa .The highly expression of PRDX3 could lead to intracellular reactive oxygen species ( ROS) increased and excessive ROS would attack the intracellular molecules , leading to DNA damage , lipid peroxidation and protein oxidation , which promote tumor cell growth .It may be combined with ROS levels to diagnosis PCa early and judge the degree of the malignancy of PCa .
