目的:观察归肺理气方对痰热壅肺证慢性阻塞性肺疾病(COPD)大鼠血清与肺组织基质金属蛋白酶-9(MMP-9)和基质金属蛋白酶组织抑制剂-1(TIMP-1)的影响。方法将61只SD大鼠随机分为7组(正常组、正常+中药中剂量组、模型组、中药低剂量组、中药中剂量组、中药高剂量组、泼尼松组),采用香烟熏吸加气管内注入脂多糖(LPS)法建立大鼠痰热壅肺证COPD模型。予归肺理气方高、中、低3个浓度及强的松灌胃干预28d,检测血清与肺组织MMP-9和TIMP-1含量。结果模型组大鼠血清与肺组织的MMP-9和TIMP-1含量以及MMP-9/TIMP-1值较正常组显著增加(P<0.05),中药中、高剂量组与泼尼松组的MMP-9量以及MMP-9/TIMP-1值较模型组显著下降(P<0.05)。结论归肺理气方可调节痰热壅肺证慢性阻塞性肺疾病大鼠的MMP-9/TIMP-1失衡,可能有一定的改善COPD气道重塑作用。
目的:觀察歸肺理氣方對痰熱壅肺證慢性阻塞性肺疾病(COPD)大鼠血清與肺組織基質金屬蛋白酶-9(MMP-9)和基質金屬蛋白酶組織抑製劑-1(TIMP-1)的影響。方法將61隻SD大鼠隨機分為7組(正常組、正常+中藥中劑量組、模型組、中藥低劑量組、中藥中劑量組、中藥高劑量組、潑尼鬆組),採用香煙熏吸加氣管內註入脂多糖(LPS)法建立大鼠痰熱壅肺證COPD模型。予歸肺理氣方高、中、低3箇濃度及彊的鬆灌胃榦預28d,檢測血清與肺組織MMP-9和TIMP-1含量。結果模型組大鼠血清與肺組織的MMP-9和TIMP-1含量以及MMP-9/TIMP-1值較正常組顯著增加(P<0.05),中藥中、高劑量組與潑尼鬆組的MMP-9量以及MMP-9/TIMP-1值較模型組顯著下降(P<0.05)。結論歸肺理氣方可調節痰熱壅肺證慢性阻塞性肺疾病大鼠的MMP-9/TIMP-1失衡,可能有一定的改善COPD氣道重塑作用。
목적:관찰귀폐리기방대담열옹폐증만성조새성폐질병(COPD)대서혈청여폐조직기질금속단백매-9(MMP-9)화기질금속단백매조직억제제-1(TIMP-1)적영향。방법장61지SD대서수궤분위7조(정상조、정상+중약중제량조、모형조、중약저제량조、중약중제량조、중약고제량조、발니송조),채용향연훈흡가기관내주입지다당(LPS)법건립대서담열옹폐증COPD모형。여귀폐리기방고、중、저3개농도급강적송관위간예28d,검측혈청여폐조직MMP-9화TIMP-1함량。결과모형조대서혈청여폐조직적MMP-9화TIMP-1함량이급MMP-9/TIMP-1치교정상조현저증가(P<0.05),중약중、고제량조여발니송조적MMP-9량이급MMP-9/TIMP-1치교모형조현저하강(P<0.05)。결론귀폐리기방가조절담열옹폐증만성조새성폐질병대서적MMP-9/TIMP-1실형,가능유일정적개선COPD기도중소작용。
Objective To observe effect of GuifeiLiqiRecipe on the expression of MMP-9 and TIM P-1 in blood serum and lung tissue of the "phlegm obstruct the lung" syndromes of COPD rats. Methods 61SD rats were randomly divided into seven groups (normal group, normal+traditional Chinese medicine in dose of middle group, traditional Chinese medicine in dose of low, middle, high group, prednisone group). The rat model of COPD was established by using the method of cigarette smoking combined with intratracheal injection of lipopolysaccharide (LPS). The content of MMP-9 and TIMP-1 in blood serum and lung tissue were detected. Results The content of MMP-9, TIMP-1 and MMP-9/TIMP-1 in blood serum and lung tissue of the model group increased significantly compared with those of the normal group (P < 0.05). The content of MMP-9 and MMP-9/TIMP-1 of the traditional Chinese medicine in dose of middle, high group and prednisone group decreased significantly compared with those of the model group (P < 0.05). Conclusion GuifeiLiqiRecipe can adjust the MMP-9/TIMP-1 imbalance on the phlegm obstruct the lung syndromes of COPD rats. It perhaps has partial effect on improving air duct remodeling of COPD.
