目的:通过血小板衍生生长因子(PDGF-B)、表皮生长因子(EGF)、转化生长因子-a(TGF-a)和8-Br-cAMP共培养对多囊卵巢综合征(PCOS)患者离体卵巢黄素化颗粒细胞雌二醇(E2)生成的影响,探讨 PDGF-B 在多囊卵巢形成中的作用.方法:收集体外受精-胚胎移植(IVF-ET)时PC0S患者的卵巢黄素化颗粒细胞进行体外原代培养,在培养0、48、120 h时以睾酮(10-7mol/L)作为底物,分别添加剂量相同的(10μg/L)PDGF-B和TGF-a(A组);PDGF-B和8-Br-cAMP (B组);PDGF-B和EGF(C组);PDGF-B、TGF-a和EGF(D组);PDGF-B、EGF、TGF-a和8-Br-cAMP (E组)于无血清培养液中,培养3 h,收集培养上清,用放射免疫法测定E2含量.以不加任何细胞因子为空白对照.结果:与空白对照比较,PCOS患者颗粒细胞经四种因子共同培养,A组在Oh和120h E2分泌量显著增加(P <0.05);B组在48h和120h E2分泌量显著增加(P<0.05);C组在0h和120h显著增加E2分泌量(P <0.05);D组在120h显著增加E2分泌量(P <0.05);E组在48h和120h E2分泌量显著增加(P <0.05).结论:PDGF-B、TGF-a、EGF和8-Br-cAMP共同作用,增加PCOS患者颗粒细胞E2的分泌,PDGF-B可能与多囊卵巢的形成有关.

Objective:To investigate the roles of PDGF-B in polycystic ovary formation by co-cultivation of platelet-derived growth factor (PDGF-B), epidermal growth factor (EGF), transforming growth factor-a (TGF-a) and 8-Br-cAMP on luteinized granulosa cell estradiol (E2) in polycystic ovary syndrome (PCOS) patients. Method: The luteinized granulesa cells were prepared from follicular aspirates during in vitro fertilization and embryo transfer (IVF-ET) , and pre—incubated for 0h,48h,120h.using testosterone (10-7mol/L) as substrate with or without the same amount (10μg/L) of PDGF-B and TGF-a (group A), PDGF-B and 8-Br-cAMP(group B), PDGF-B and EGF(group C), PDGF-B、TGF-aand EGF(group D), PDGF-B、EGF、TGF-aand 8-Br-cAMP (group E)in serum-free medium, respectively. The end of the culture,the medium were collected for the measurements 0f oestrediol by RIA. The cells were lysed with 50ul 0.1 mol/L sodium hydroxide and their protein concentration were assayed by CBBG一250/BSA in order to correct E2,and luteinized granulosa cells without added any cytokines as a control group. Result:Compared with a control group, PCOS patients granulosa cells E2 secretion amount at 0h and 120h were significantly increased (P<0.05) in group A;at 48h and 120h E2 secretion amount were significantly increased (P<0.05) in group B;at 0h and 120h E2 secretion were increased significantly (P<0.05) in group C; at 120h E2 secretion were significantly increased in group D (P <0.05); and at 48h and 120h E2 secretion were increased significantly (P<0.05) in group E. Conclusion:PDGF-B、TGF-a、EGF and 8-Br-cAMP together increase E2 secretion of PCOS patients, PDGF-B may be associated with polycystic ovary formation.
