利用阳极氧化法在纯 Ti 表面制得 TiO2纳米管,随后用冻干法在纳米管中负载 microRNA 制得复合涂层。利用 SEM、TEM和荧光显微镜对 TiO2/microRNA复合涂层进行表征,并对 microRNA 的释放行为以及其进入成骨细胞后的功效进行分析。结果表明,microRNA分子成功负载在 TiO2纳米管中,并在12 h内持续稳定释放投递,荧光照片表明投递的microRNA成功进入成骨细胞,ALP 活性测定进一步表明进入细胞中 microRNA促进了成骨分化。
利用暘極氧化法在純 Ti 錶麵製得 TiO2納米管,隨後用凍榦法在納米管中負載 microRNA 製得複閤塗層。利用 SEM、TEM和熒光顯微鏡對 TiO2/microRNA複閤塗層進行錶徵,併對 microRNA 的釋放行為以及其進入成骨細胞後的功效進行分析。結果錶明,microRNA分子成功負載在 TiO2納米管中,併在12 h內持續穩定釋放投遞,熒光照片錶明投遞的microRNA成功進入成骨細胞,ALP 活性測定進一步錶明進入細胞中 microRNA促進瞭成骨分化。
이용양겁양화법재순 Ti 표면제득 TiO2납미관,수후용동간법재납미관중부재 microRNA 제득복합도층。이용 SEM、TEM화형광현미경대 TiO2/microRNA복합도층진행표정,병대 microRNA 적석방행위이급기진입성골세포후적공효진행분석。결과표명,microRNA분자성공부재재 TiO2납미관중,병재12 h내지속은정석방투체,형광조편표명투체적microRNA성공진입성골세포,ALP 활성측정진일보표명진입세포중 microRNA촉진료성골분화。
TiO2 nanotubes was prepared by anodic oxidation method on pure Ti surface,then the microRNA was loaded in the nanotubes by lyophilization to build composite coating.The TiO2/microRNA composite coating was characterized by SEM,TEM and fluorescence microscope.The release behavior of microRNA and its furthrer function was analysised.The results showed that microRNA successfully deposited in TiO2 nanotubes, and sustained release delivery for 1 2 h.Fluorescent images indicated that the microRNA was transfected into osteoblasts,ALP activity assay further indicated that the microRNA promoted the osteogenic differentiation.
