目的:研究乌芪通络胶囊对糖尿病周围神经病变大鼠背根神经节中JNK1/2和p38 MAPK蛋白表达的影响,从氧化应激的角度探讨乌芪通络胶囊治疗糖尿病周围神经病变的机制。方法:实验用雄性SD大鼠80只,随机选择14只为正常对照组,其余66只用来造模。后者经腹腔注射链脲佐菌素( STZ),成功造模51只。将造模成功的大鼠随机分为模型组、弥可保组及乌芪通络胶囊组,每组17只。弥可保组和乌芪通络胶囊组连续给药8周,对照组和模型组正常饮食8周,将8周后的四组大鼠处死,取背根神经的L4/L5神经节,提取总蛋白,采用Western杂交的方法检测背根神经节中JNK1/2和p38 MAPK蛋白的表达水平。结果:模型组JNK1/2和p38 MAPK蛋白的表达比正常组显著增高( P<0.05)。乌芪通络胶囊组和弥可保组JNK1/2和p38MAPK蛋白表达较模型组显著降低,差异有统计学意义(P<0.05)。结论:乌芪通络胶囊可降低大鼠背根神经节中的JNK1/2和p38 MAPK蛋白的表达,这可能是乌芪通络胶囊从氧化应激方面治疗和缓解糖尿病周围神经病变各种症状的机制之一。
目的:研究烏芪通絡膠囊對糖尿病週圍神經病變大鼠揹根神經節中JNK1/2和p38 MAPK蛋白錶達的影響,從氧化應激的角度探討烏芪通絡膠囊治療糖尿病週圍神經病變的機製。方法:實驗用雄性SD大鼠80隻,隨機選擇14隻為正常對照組,其餘66隻用來造模。後者經腹腔註射鏈脲佐菌素( STZ),成功造模51隻。將造模成功的大鼠隨機分為模型組、瀰可保組及烏芪通絡膠囊組,每組17隻。瀰可保組和烏芪通絡膠囊組連續給藥8週,對照組和模型組正常飲食8週,將8週後的四組大鼠處死,取揹根神經的L4/L5神經節,提取總蛋白,採用Western雜交的方法檢測揹根神經節中JNK1/2和p38 MAPK蛋白的錶達水平。結果:模型組JNK1/2和p38 MAPK蛋白的錶達比正常組顯著增高( P<0.05)。烏芪通絡膠囊組和瀰可保組JNK1/2和p38MAPK蛋白錶達較模型組顯著降低,差異有統計學意義(P<0.05)。結論:烏芪通絡膠囊可降低大鼠揹根神經節中的JNK1/2和p38 MAPK蛋白的錶達,這可能是烏芪通絡膠囊從氧化應激方麵治療和緩解糖尿病週圍神經病變各種癥狀的機製之一。
목적:연구오기통락효낭대당뇨병주위신경병변대서배근신경절중JNK1/2화p38 MAPK단백표체적영향,종양화응격적각도탐토오기통락효낭치료당뇨병주위신경병변적궤제。방법:실험용웅성SD대서80지,수궤선택14지위정상대조조,기여66지용래조모。후자경복강주사련뇨좌균소( STZ),성공조모51지。장조모성공적대서수궤분위모형조、미가보조급오기통락효낭조,매조17지。미가보조화오기통락효낭조련속급약8주,대조조화모형조정상음식8주,장8주후적사조대서처사,취배근신경적L4/L5신경절,제취총단백,채용Western잡교적방법검측배근신경절중JNK1/2화p38 MAPK단백적표체수평。결과:모형조JNK1/2화p38 MAPK단백적표체비정상조현저증고( P<0.05)。오기통락효낭조화미가보조JNK1/2화p38MAPK단백표체교모형조현저강저,차이유통계학의의(P<0.05)。결론:오기통락효낭가강저대서배근신경절중적JNK1/2화p38 MAPK단백적표체,저가능시오기통락효낭종양화응격방면치료화완해당뇨병주위신경병변각충증상적궤제지일。
Objective:In order to explore the mechanism of Wuqi Tongluo capsule on diabetic peripheral neuropathy ( DPN) from oxidative stress aspect , we observed the protein expression of JNK 1/2 and p38MAPK in dorsal root ganglion (DRG) of DPN rats.Methods:80 male SD rats were randomly divided into two groups:model group(66 rats) and nor-mal group(14 rats) .The model group was molded with STZ Intraperitoneal injection , and obtained 51 successful mod-els.The DPN model group was randomly divided into model group , Wuqi Tongluo capsule group and Methycobal group , with 17 rats in each group .After eight weeks ’ administration , extract total protein from L4/L5 ganglion of rats ’ DRG. The JNK1/2 and p38 MAPK expression levels in DRG of each group were determined by western blot .Results:The pro-tein expression of JNK1/2 and p38MAPK in model group increased obviously than that in the normal group (P<0.05). Wuqi Tongluo capsule group and Methycobal group obviously decreased compared with the model group , and the differ-ence was statistically significant ( P<0 .05 ) .Conclusion:Wuqi Tongluo capsule can decrease the protein expression of JNK1/2 and p38MAPK in DPN rats, which may be one of mechanisms of Wuqi Tongluo capsule in the treatment of dia-betic peripheral neuropathy from oxidative stress aspect .
