应用实时荧光定量PCR技术,通过室内营养钵土壤人工接种粉红粘帚霉67-1菌株,研究土壤温度、pH、含水率对生防菌粉红粘帚霉在土壤中消长动态的影响。结果表明:在不同温度、含水率和酸碱度的土壤中,菌株67-1种群动态变化曲线基本一致,接种5 d前土壤中67-1拷贝数明显减少,10 d后拷贝数逐渐增加,20 d时达到最高,并维持在这一水平。土壤因子对菌株67-1的定殖量具有显著影响,接种相同时间30℃时土壤中菌株67-1拷贝数增加最快,其次是20℃,10℃最低;土壤pH值为5.0时拷贝数最高, pH为8.0时拷贝数较低,pH为4.0时拷贝数最低;土壤含水率为15%时菌株67-1拷贝数高于含水率25%,但二者差异不显著,表明菌株67-1在土壤中的扩增受土壤含水率影响较小。

By using real-time fluorescence quantitative PCR method, we conducted the effects of soil temperature, pH and moisture on population dynamics of Clonostachys rosea 67-1 in greenhouse. The results showed that the curves of population dynamics of 67-1 were consistent in different soil temperature, moisture and pH. The copy numbers of the fungus decreased obviously after inoculated for 5 days, and increased after 10 days. The greatest copy numbers were obtained in 20 days, and the fungal colonization maintained at the high level for a period of time. Soil factors had obvious effects on 67-1. The increasing rate of number of 67-1 copies was the highest under 30℃, followed by 20℃. The lowest amount of the biocontrol fungus was recovered in the soil treated under 10℃. When soil pH value was adjusted to 5.0, the copy number of 67-1was the highest, while the soil with pH 4.0 or pH 8.0 was not suitable for 67-1 proliferation. The copy numbers were higher in soil with the moisture of 25%than that with the moisture of 15%, however, there was no significant difference between them, which indicated that soil moisture did not significantly affecte colonization of C. rosea 67-1.
