以鸡蛋清为原料,采用正交实验分别确定盐析法与离子交换法两种提取方法的最佳条件,同时采用比浊法测定酶活力。确定了盐析法从蛋清中提取溶菌酶的最佳条件是:加入蛋清滤液体积5%的氯化钠;结晶的温度4℃;结晶的时间为4天;离子交换法的最佳吸附条件为:蛋清树脂比5∶1,pH 7.0,搅拌吸附时间为4 h。离子交换法所得酶的产量与活力均高于盐析法。

Using egg white as material,enzyme activity as target,the alting-out method and ion exchange extraction methods were studied by orthogonal experiment design.The optimal process conditions for extraction lysozyme were adding 5% sodium chloride,crystallization temperature 4 ℃,and crystallization time for 4 days.The best conditions of Ion exchange were egg white resin ratio 5:1,pH 7.0,and stirred adsorption time for 4 h.The output and enzyme activity of enzyme derived from ion exchange was higher than that got from the salting-out method.
