实验研究了30日龄肉鸡按55.38mg/Kg.体质量剂量灌服10%硫酸锰溶液后不同时间采血,应用石墨炉原子吸收分光光度法测定血药浓度,经微机处理得出血药浓度及药动学参数,血药浓度-时间曲线符合一级吸收一室开放模型,最佳药时方程:C=80.8013(e-0.0709t-e-0.4402t)。主要药动学参数:t1/2Ka为1.5747hrs,t1/2Ke为9.7698hrs,tp为5.6617hrs,Cmax为48.3581ug/L,AUC为967.9305(μg/L).h。根据单剂量给药参数,计算出多剂量给药参数。τ定为12h,(C∝)max为95.997μg/L,c为80.6609μg/L,(C∝)min为4.0668μg/L,D*为3.2794mg/Kg.BW,D0为1.8789mg/Kg.BW,R为1.7454。

The dosage of 10% manganese sulfate 55.38 mg/Kg · BW was gavaged to 30--d--old broiler, at the different time and the determination of trace element manganese by graphite furnace atomic absorption spectrophotometry by treating microcomputer, then calculate corresponding pharmacokinetic parameter of blood drug concentration. The results were showed as follow: the characteristics of time--concentration curves conforms to the first order absorption and one compartment open mod- el, the theoretical equation was C = 80. 8013 (e-0.0709 t--e-0.4402t ) , the main parameters: the half--life of absorption (T1/zKa) was 1. 5747 hrs, the half--life elimination (T1/2Ke) was 9. 7698 hrs , the time of reaching maximum concentration (Tp) was 5. 6617 hrs, the area under the curve (AUC) was (μg/L) · h. The parameters of multiple doses were calculated for clinic treat based on steady state plasma eoncentration the kinetic parameters of the single dose, dosing internal(r) was 12 hrs, maximum steady state plasma concentration [(Coc)max] was 95. 997μg/L, average plasma concentration (c) was 80. 6609/1g/L, minimum steady state plasma concentration C(Csc )minwas 4. 0668 ttg/L, loading dose (D*) was 3. 2794 mg/Kg · BW, maintenance dose(D0) was 1. 8789 mg/Kg · BW, the accumulation factor (R) was 1. 7454.
