目的:分析新疆地区回族人群杀伤细胞免疫球蛋白样受体(KIR)基因多态性及基因型和单倍型特点,为研究 KIRs 与疾病的关系提供依据。方法采用序列特异性引物 PCR 方法(PCR‐SSP),对25例新疆回族志愿者 DNA 样本进行 KIR 基因检测和基因型及单倍型分析。结果(1)共检测已知的16个 KIR 基因,3DL3、2DL1、3DP1、2DL4、3DL2基因存在于所有25例个体,其基因频率均为100.00%;2DP1、2DL3、3DL1基因较为常见,频率分别为96.00%、96.00%、92.00%;其次2DS4、2DS1、2DL5基因频率分别为88.0%、56.00%和56.00%;2DS2、2DL2基因频率最低,均为24.00%。(2)共检出9种基因型,其中以 AF(1,2)最常见,频率为44.00%;其次为 AE(1,6)和 AH(5,2)型较常见,频率为24.0%和8.00%;而 C(5,3)、I(5,8)、P(2,17)和 Q(9,5)型较少见,频率均为4.00%;另有1例基因型(HNXJ1),根据现有相关文献不能对应分析基因型和单倍型,提示可能存在新的基因型。(3)共检出9种单倍型,最常见单倍型1(n =17)占35.4%和单倍型2(n =14)占29.2%,其次为单倍型6(n =6)占12.5%和单倍型5(n =5)占10.4%。结论新疆地区回族人群 KIR 基因具有不同民族人群的共性,也有自身的基因型和单倍型分布特征。

목적:분석신강지구회족인군살상세포면역구단백양수체(KIR)기인다태성급기인형화단배형특점,위연구 KIRs 여질병적관계제공의거。방법채용서렬특이성인물 PCR 방법(PCR‐SSP),대25례신강회족지원자 DNA 양본진행 KIR 기인검측화기인형급단배형분석。결과(1)공검측이지적16개 KIR 기인,3DL3、2DL1、3DP1、2DL4、3DL2기인존재우소유25례개체,기기인빈솔균위100.00%;2DP1、2DL3、3DL1기인교위상견,빈솔분별위96.00%、96.00%、92.00%;기차2DS4、2DS1、2DL5기인빈솔분별위88.0%、56.00%화56.00%;2DS2、2DL2기인빈솔최저,균위24.00%。(2)공검출9충기인형,기중이 AF(1,2)최상견,빈솔위44.00%;기차위 AE(1,6)화 AH(5,2)형교상견,빈솔위24.0%화8.00%;이 C(5,3)、I(5,8)、P(2,17)화 Q(9,5)형교소견,빈솔균위4.00%;령유1례기인형(HNXJ1),근거현유상관문헌불능대응분석기인형화단배형,제시가능존재신적기인형。(3)공검출9충단배형,최상견단배형1(n =17)점35.4%화단배형2(n =14)점29.2%,기차위단배형6(n =6)점12.5%화단배형5(n =5)점10.4%。결론신강지구회족인군 KIR 기인구유불동민족인군적공성,야유자신적기인형화단배형분포특정。
Objective To analyze the gene polymorphism genotype and haplotype characteristics of killer cell immunoglobulin‐like receptors (KIR) in Xinjiang Hui people and to lay the basis for the study of the KIRs‐related disease .Methods Sequence‐specific primer PCR method (PCR‐SSP) was adopted to conduct the KIR gene detection , genotype and haplotype analysis on 25 cases randomly‐selected samples of Xinjiang Hui volunteers .Results (1) 16 known KIR genes were detected :3DL3 ,2DL1 ,3DP1 ,2DL4 , 3DL2 genes existed in all 25 individual cases with 100% gene frequency ;2DP1 ,2DL3 ,3DL1 genes were common with gene frequency of 96 .00% ,96 .00% ,92 .00% ;followed by 2DS4 ,2DS1 ,2DL5 with gene frequency of 88 .0% ,56 .00% ,56 .00% ;2DS2 of 2DL2 gene had minimum gene frequency of 24 .00% .(2) eight genotypes were detected :AF (1 ,2) had the most common frequency of 44 .00% ;followed by the type of AE (1 ,6) and AH (5 ,2) with the common frequency of 24 .0% and 8 .00 % ;C (5 ,3) ,I (5 ,8) ,P (2 ,17) ,and Q (9 ,5) had the lowest frequency of 4 .00% .There existed one genotype (HNXJ1) case that didn′t correspond to the analysis of the genotype and haplotype according to the literature , which suggested a new genotype ;(3) 9 haplotypes were detected :the most common haplotype 1 (n = 17) ac‐counted for 35 .4% ,and haplotype 2 (n = 14) accounted for 29 .2% ,followed by haplotype 6 (n = 6) ac‐counted for 12 .5% and haplotype 5 (n = 5) accounted for 10 .4% .Conclusion KIR gene in Xinjiang Hui people has commonalities of different ethnic groups ,and also has its own genotype and haplotype distribu‐tion characteristics .
