目的:考察全氟辛烷磺酸(PFOS)暴露致大鼠肝损伤的作用及机制。方法48只 Wistar 大鼠随机分为对照组、低剂量组和高剂量组,分别给予含0、4、8 mg/kg PFOS 饲料染毒3个月。高效液相/质谱法检测大鼠血清和肝脏中 PFOS 含量;ELISA 法测定各组丙氨酸转氨酶(ALT )、谷草转氨酶(AST )、碱性磷酸酶(ALP)、层粘连蛋白(LN)和羟脯氨酸(Hyp)含量变化。紫外分光光度法测定肝组织匀浆中单胺氧化酶(MAO)、超氧化物歧化酶(SOD)、谷胱甘肽(GSH)、琥珀酸脱氢酶(SDH)活性变化。结果 PFOS 暴露致大鼠血清及肝脏中 PFOS 浓度显著增加,肝脏与血清中 PFOS 比值分别为(4.22±0.99)和(3.33±0.75)(P <0.05)。 PFOS 暴露后大鼠肝脏重量和脏器系数显著增高(P <0.05),组织切片显示暴露造成肝肿大和脂肪化发生。染毒组大鼠血清 ALT 、ALP 和 AST均显著升高(P <0.05);Hyp 和 LN 在 PFOS 高剂量暴露组显著增高(P <0.05)。染毒组大鼠肝细胞中 MAO 和GSH 水平高于对照组,而 SDH 和 SOD 活力较对照组显著下降(P <0.05)。结论 PFOS 暴露可能通过诱发氧化应激造成大鼠肝损伤。
目的:攷察全氟辛烷磺痠(PFOS)暴露緻大鼠肝損傷的作用及機製。方法48隻 Wistar 大鼠隨機分為對照組、低劑量組和高劑量組,分彆給予含0、4、8 mg/kg PFOS 飼料染毒3箇月。高效液相/質譜法檢測大鼠血清和肝髒中 PFOS 含量;ELISA 法測定各組丙氨痠轉氨酶(ALT )、穀草轉氨酶(AST )、堿性燐痠酶(ALP)、層粘連蛋白(LN)和羥脯氨痠(Hyp)含量變化。紫外分光光度法測定肝組織勻漿中單胺氧化酶(MAO)、超氧化物歧化酶(SOD)、穀胱甘肽(GSH)、琥珀痠脫氫酶(SDH)活性變化。結果 PFOS 暴露緻大鼠血清及肝髒中 PFOS 濃度顯著增加,肝髒與血清中 PFOS 比值分彆為(4.22±0.99)和(3.33±0.75)(P <0.05)。 PFOS 暴露後大鼠肝髒重量和髒器繫數顯著增高(P <0.05),組織切片顯示暴露造成肝腫大和脂肪化髮生。染毒組大鼠血清 ALT 、ALP 和 AST均顯著升高(P <0.05);Hyp 和 LN 在 PFOS 高劑量暴露組顯著增高(P <0.05)。染毒組大鼠肝細胞中 MAO 和GSH 水平高于對照組,而 SDH 和 SOD 活力較對照組顯著下降(P <0.05)。結論 PFOS 暴露可能通過誘髮氧化應激造成大鼠肝損傷。
목적:고찰전불신완광산(PFOS)폭로치대서간손상적작용급궤제。방법48지 Wistar 대서수궤분위대조조、저제량조화고제량조,분별급여함0、4、8 mg/kg PFOS 사료염독3개월。고효액상/질보법검측대서혈청화간장중 PFOS 함량;ELISA 법측정각조병안산전안매(ALT )、곡초전안매(AST )、감성린산매(ALP)、층점련단백(LN)화간포안산(Hyp)함량변화。자외분광광도법측정간조직균장중단알양화매(MAO)、초양화물기화매(SOD)、곡광감태(GSH)、호박산탈경매(SDH)활성변화。결과 PFOS 폭로치대서혈청급간장중 PFOS 농도현저증가,간장여혈청중 PFOS 비치분별위(4.22±0.99)화(3.33±0.75)(P <0.05)。 PFOS 폭로후대서간장중량화장기계수현저증고(P <0.05),조직절편현시폭로조성간종대화지방화발생。염독조대서혈청 ALT 、ALP 화 AST균현저승고(P <0.05);Hyp 화 LN 재 PFOS 고제량폭로조현저증고(P <0.05)。염독조대서간세포중 MAO 화GSH 수평고우대조조,이 SDH 화 SOD 활력교대조조현저하강(P <0.05)。결론 PFOS 폭로가능통과유발양화응격조성대서간손상。
Objective To study the effect of perfluorooctane sulfonate (PFOS) exposure on rat liver .Meth‐ods Rats were exposed with 0 mg/kg ,4 mg/kg and 8 mg/kg PFOS food for 3 months .Rats were sacri‐ficed and PFOS concentration was determined by HPLC‐LC/MS ,histological changes of the liver tissue were examined as well .The contents including ALT ,AST ,ALP ,LN and Hyp in serum were determined by ELISA kits .Besides ,MAO ,SOD ,GSH and SDH were detected by the colorimetric method .Results It showed that PFOS concentration increased after exposure and the ration of PFOSliver /serum was (4 .22 ± 0 .99) and (3 .33 ± 0 .75) respectively (P < 0 .05) .Furthermore ,PFOS exposure resulted in a significant hepatic injury with cell edema and diffuse steatosis in both groups (P < 0 .05) .Compared with the control , ALT ,ALP and AST increased significantly ,especially in the high dose group (P < 0 .05) .Additionally , Hyp and LN ,the indicators of liver fibrosis ,significantly increased (P < 0 .05) .Moreover ,PFOS exposure increased the levels of MDA and GSH while reduced the levels of SDH and SOD (P < 0 .05) .Conclusion There was a significant injury effect with rat liver after PFOS exposure .The mechanism of action is related to oxidative stress .
