目的:探讨小睑裂综合征(BPES)患者血液中FOXL2,SOX14及BPESC13个基因mRNA的相对表达水平与BPES的相关性,以进一步研究可能存在的发病机制。方法收集小睑裂综合征患者( A组30例)和正常人( B组30例)的外周静脉血液,TRIzol提取总RNA,反转录为cDNA。应用荧光定量PCR技术检测FOXL2, BPESC1,SOX14的相对表达量,采用配对比较 t 检验法和 Livak(2-ΔΔCt)法分析两组数据及其差异。结果FOXL2基因mRNA的在A,B两组间ΔCt差异具有统计学意义(P=0.023<0.05),且A组大于B组;BPESC1差异亦有统计学意义(P=0.008<0.05),且A组小于B组;SOX14差异无统计学意义(P=0.157>0.05)。可以认为A组FOXL2基因的表达水平低于B组,BPESC1基因的表达水平高于B组,SOX14基因两组表达无差别。结论 FOXL2,BPESC1与BPES具有一定相关性,FOXL2低表达以及BPESC1的高表达可能与BPES的发病相关。

Objective To detect mRNA expression of FOXL2,SOX14 and BPESC1 from BPES patients and define the correla-tion between the exoression and BPES ,in order to expore the possible pathogenesis of BPES .Methods The peripheral venous blood was col-lected from 30 cases of BFPS patients(Group A)and 30 cases of normal individuals(Group B).RNA was extracted by TRIzol and then was transcripted to cDNA reversely .Real -time fluorescence quantitative PCR was applied to detect the relative mRNA expression levels of FOXL2,SOX14 and BPESC1 in blood from Group A and Group B .Two paired sample test and Livak (2 -ΔΔCt ) method were used to analyze the differences between two data sets .Results Between groups A and B,PFOXL2 =0.023 <0.05 and Group A was higher than Group B;PBPESC1 =0.008<0.05 and Group A was lower than Group B;PSOX14 =0.157>0.05,there was no statistical differences .It indicated that FOXL2 expression in Group A was lower than that in Group B ,while BPESC1 expression in Group A was higher than that in Group B .Con-clusion FOXL2 and BPESC1 are associated with BPES ,and low expression of FOXL2 and high expression of BPESC 1 might be related to BPES pathogenesis .
