目的:研究血管紧张素Ⅱ(AⅡ)和肾素活性(PRA)在不同温度条件下对测定结果的影响。方法90例标本分成三组,第一组采血后4℃低温离心。第二组室温放置2h后4℃低温离心。第三组在4℃冰箱放置2h后4℃低温离心。离心后三组都在30min内上机测定结果。另外,将20例标本分别放入4℃和-20℃冰箱,取0d、1d、2d、3d、7d、14d为时间点,测定结果保持稳定的天数。结果在分组实验中第二组与第一组相比AⅡ结果降低(20.21±10.98)%,PRA结果降低(25.6±9.76)%(P<0.05),第三组与第一组相比AⅡ结果降低(1.60±0.85)%,PRA结果降低(3.3±1.18)%(P>0.05),在4℃环境中,AⅡ与PRA前3天结果没有显著性差异,P>0.05。3天到14天有显著性差异,P<0.05。在-20℃环境中AⅡ标本在14天内6个时间点的结果没有显著性差异,P>0.05。 PRA在7天内的结果无统计学差异,P>0.05。而7天到14天有显著性差异,P<0.05。结论 AⅡ和PRA对温度敏感,室温放置能使其结果偏低,而在低温环境中能够保持结果稳定。建议AⅡ和PRA标本的运送与预处理都应该在低温中进行,并制定详细的操作规程确保在日常工作中流程的可靠。

Objective To study the concentration changes of plasma angiotensinⅡ(AⅡ) and renin activity (PRA) in different temperature conditions. Methods Ninety blood samples were collected from 30 patients and divided into 3 groups. In the first group, the procedure followed the handling protocol provided by the manufacturers for the AⅡand PRA kits precisely. The other two groups kept at room temperature and 40C for 2 hours respectively, and then processed samples by the handling protocol the first group followed. In addition, other 20 specimens were separately stored for 1, 2, 3, 7 and 14 days at 4°C and -20°C to evalu-ate the stability of AⅡand PRA. Results When blood samples were kept at room temperature for 2 hours, the PRA and AⅡde-creased significantly (P<0.05), but both unchanged if samples were placed at 4°C for the same time period. During storage at 4°C, the PRA and AⅡgot no significant changes for 3 days. After 3 days, PRA and AⅡdecreased rapidly. When put into the-20°C en-vironment, the PRA and AⅡwere stabilized for a week. Conclusion The AⅡand PRA were sensitive to temperature, and de-creased quickly in a room temperature. But a low temperature storage condition would remain them in a stable state. So it is neces-sary to apply well-defined pre-analytical procedures for AⅡand PRA measurements.
