采用HPLC的模式,分别研究温敏性色谱固定相对不同极性的类固醇、多肽等物质的驻留性能,考察物质极性等性质对其驻留性能的影响。实验发现,多肽样品随疏水性的增强,保留时间发生不同程度的延后现象,疏水性越强延后程度越明显。同时发现,溶液中的高离子强度可以增强蛋白质和多肽等生物大分子与疏水性固定相介质之间的疏水作用。
採用HPLC的模式,分彆研究溫敏性色譜固定相對不同極性的類固醇、多肽等物質的駐留性能,攷察物質極性等性質對其駐留性能的影響。實驗髮現,多肽樣品隨疏水性的增彊,保留時間髮生不同程度的延後現象,疏水性越彊延後程度越明顯。同時髮現,溶液中的高離子彊度可以增彊蛋白質和多肽等生物大分子與疏水性固定相介質之間的疏水作用。

Poly(N-isopropylacrylamide)(PNIPAAm) grafted to(aminopropyl)silica by an surface two-bond coupling was used as an HPLC packing material.The surface properties and functions of the stationary phase of some steroids and some peptides were controlled by external temperature.Isocratic elution by aqueous mobile phase of alone was the basis for separation of peptides.The elution behaviors of peptides with different hydrophobicity and isoelectric point were controlled by the temperature changes without organic solvents in the mobile phase.Separation was optimized just by changing the temperature,instead of changing the mobile phase composition.With increasing temperature,increased interaction between solutes and surfaces of the stationary phases was observed.
