对大孔吸附树脂富集纯化虎舌红(Ardisia mamillata)总黄酮的最佳工艺参数进行了研究,并比较了4种大孔树脂对虎舌红总黄酮的吸附与解吸效果,从中筛选出适合分离纯化虎舌红总黄酮的树脂,同时对其吸附和解吸条件进行了探讨.结果表明,AB-8型为较理想树脂,其纯化总黄酮的最优工艺条件为:上样液质量浓度为0.34mg/mL,吸附流速为2.0mL/min,吸附pH值为5.0,洗脱剂为3BV(90mL)体积分数70%的乙醇,解吸流速为1.50mL/min.经AB-8型树脂纯化后,总黄酮纯度由原来的8.92%提高到67.79%,提高了7.6倍.

The optimal conditions of purification of total flavonoids from Ardisia mamillata with macro- porous resin were studied, i.e. the absorption and desorption properties of four kinds of macroporous resin. Results showed that AB-8 macroporous resin had the best adsorption capacity under following technic condi- tions: the initial concentration, flow velouty, pH value and desovption flow velouty were at 0. 34 mg/mL, 2. 0 mL/min, 5.0 and 1.50 mL/min, respectively, with 3BV 70% ethanol as washiny solution. The purity of flavonoids was from 8.92% to 67.79%, which was 7.6 times higher than that of the original sample.
