为提高粳稻抗性愈伤分化率和遗传转化率,以农杆菌介导遗传转化得到的水稻成熟胚抗性愈伤(Hyg)为材料,研究了影响粳稻成熟胚抗性愈伤组织再生频率的因素。结果表明分化培养基添加0.5~25 mg.L-1的CuSO4.5H2O,增加分化培养基中的琼脂浓度,分化前对抗性愈伤进行高渗处理,采用6-BA和KT两步分化法和干燥除菌法均能提高抗性愈伤组织的分化率。

위제고갱도항성유상분화솔화유전전화솔,이농간균개도유전전화득도적수도성숙배항성유상(Hyg)위재료,연구료영향갱도성숙배항성유상조직재생빈솔적인소。결과표명분화배양기첨가0.5~25 mg.L-1적CuSO4.5H2O,증가분화배양기중적경지농도,분화전대항성유상진행고삼처리,채용6-BA화KT량보분화법화간조제균법균능제고항성유상조직적분화솔。
To improve differentiation and transformation ratio,resistant calli(Hyg) in mature embryo of Japonica rice,obtained through genetic transformation by Agrobacterium associated method,were used to study the influence factors of regeneration frequency. The results indicated that supplement with 0.5~25 mg·L-1 CuSO4·5H2O and strengthening agar concentration in differentiation medium could improve significantly differentiation rate of resistant calli from Japonica rice,as well as other measures including high osmotic pressure treatment of calli,the two-step regeneration with 6-BA and KT in turn and the dessication treatment to eliminate Agrobacterium.
