对39份马铃薯晚疫病菌菌株的SSR扩增产物进行聚丙烯酰胺凝胶电泳后,分别用银染、溴化乙锭(EB)染色并统计清晰特征条带。实验结果表明银染分辨率高,但检测到较多的杂带,EB染色的灵敏度虽比银染差,仍能获得高质量的目的条带;理论计算的SSR目的条带的质量范围是147.5~226.7 ng,都超过了2种染色方法需要的最小理论质量。实验结果和理论分析同时表明2种染色方法皆可用于马铃薯晚疫病群体多样性的研究。

After polyacrylamide gel electrophoresis using SSR products of 39 strains isolated, silver staining and ethidium bromide (EB) staining were used to band statistic, the results show that the silver staining method lead to high resolution, but also detected more minor band, EB staining method was less sensitivity than silver staining. However,it can detect more qualified target band. In theory, calculation of SSR band quality were range from 147.5 - 226.7 ng, representing higher col- oration quality requirements then two staining methods. The experimental results and theoretical analysis also appear that the two staining methods can be used for p. infestans population study.
