Er Huang Yu Yang San%infectious skin ulcer%inflammatory factor%tumor necrosis factor%interleukin -8%rat model
目的:观察二黄愈疡散对感染性皮肤溃疡模型大鼠炎症因子、肿瘤坏死因子(tnf-α)、白介素-8(il-8)基因及蛋白表达的影响。方法实验大鼠全麻后背部脱毛区蒸汽烫40 sec,造成溃疡模型,冷却15 min 后注射大肠杆菌液,造成感染性溃疡模型。造模5 d 后切除表面痂皮,分为感染性溃疡组和非感染性溃疡组,分别给予二黄愈疡散治疗,同时分别以康复新液和生理盐水作对照。用免疫组织化学方法和 image-Pro-Plus 图像分析系统对肿瘤坏死因子(tnf-α)、白介素-8(il-8)的蛋白表达进行定量分析。结果感染性溃疡中,治疗组 il-8的水平低于康复新液组和生理盐水组(P <0.01);非感染性溃疡中,治疗组 il-8、tnf-α的水平低于康复新液组和生理盐水组(P <0.01)。结论二黄愈疡散具有明显的抗炎作用,其抗炎作用优于康复新液(P <0.01);下调 tnf-α、il-8可能是二黄愈疡散抗炎作用的重要分子机制之一。

목적:관찰이황유양산대감염성피부궤양모형대서염증인자、종류배사인자(tnf-α)、백개소-8(il-8)기인급단백표체적영향。방법실험대서전마후배부탈모구증기탕40 sec,조성궤양모형,냉각15 min 후주사대장간균액,조성감염성궤양모형。조모5 d 후절제표면가피,분위감염성궤양조화비감염성궤양조,분별급여이황유양산치료,동시분별이강복신액화생리염수작대조。용면역조직화학방법화 image-Pro-Plus 도상분석계통대종류배사인자(tnf-α)、백개소-8(il-8)적단백표체진행정량분석。결과감염성궤양중,치료조 il-8적수평저우강복신액조화생리염수조(P <0.01);비감염성궤양중,치료조 il-8、tnf-α적수평저우강복신액조화생리염수조(P <0.01)。결론이황유양산구유명현적항염작용,기항염작용우우강복신액(P <0.01);하조 tnf-α、il-8가능시이황유양산항염작용적중요분자궤제지일。
Objective To study the effects of Er Huang Yu Yang San on expressions of inflammatory factor, tumor necrosis factor (TNF-α) and interleukin -8 (IL-8) of experimental infectious skin ulcer in rats. Methods rats were ironed by steam for 40 seconds on their back hair removal zone after general anesthesia to make ulcer model, and then burn-zone cooling for 15 minutes before injection of E. coli to make infectious ulcer model. After five days of modeling, the wound surface crusts were removed, and rats were divided into infectious ulcer group and the non-infectious ulcer group. they were given treatment by er Huang Yu Yang san, at the same time rats treated with Kang fu Xin treatment group and saline group were selected as the control. the immunohistochemical methods and image-Pro-Plus image analysis system were used for quantitative analysis of protein expression of tumor necrosis factor (TNF-α) and interleukin-8 (IL-8).Result in the treatment group of infectious ulcers, the level of il-8 in treatment group was lower than that of Kang fu Xin treatment group and saline treatment group (P < 0.01). In the treatment of non-infectious ulcer, the level of IL-8 and TNF-α of Er Huang YuYang San treatment group were lower than that of Kang fu Xin treatment group and saline treatment group (P < 0.01).Conclution er Huang Yu Yang san has obvious anti-inflammatory effect, and its anti-inflammatory effect is better than that of Kangfuxin solution (P < 0.01). downregulation the level of TNF- α and IL-8 may be an important molecular mechanism of anti-inflammatory effect of Er Huang Yu Yang San.
