目的:研究胰岛素样生长因子Ⅱ( IGFⅡ)对软骨细胞基质金属蛋白酶1( MMP-1)基因表达调节的机制。方法采用体外新生大鼠股骨髁软骨细胞培养,同时加入不同剂量白介素-1β( IL-1β),使用酶联免疫吸附技术检测体外培养的关节软骨细胞上清液24h、48h、72h中基因蛋白MMP-1mRNA的表达情况,并加入IGFⅡ干预,观察IGFⅡ的效果。结果 IL-1β可增加关节软骨细胞MMP-1基因蛋白的分泌,加入IGFⅡ干预后可减少关节软骨细胞MMP-1基因蛋白的分泌。结论 IGFⅡ具有显著促进关节软骨细胞增殖的效果。

목적:연구이도소양생장인자Ⅱ( IGFⅡ)대연골세포기질금속단백매1( MMP-1)기인표체조절적궤제。방법채용체외신생대서고골과연골세포배양,동시가입불동제량백개소-1β( IL-1β),사용매련면역흡부기술검측체외배양적관절연골세포상청액24h、48h、72h중기인단백MMP-1mRNA적표체정황,병가입IGFⅡ간예,관찰IGFⅡ적효과。결과 IL-1β가증가관절연골세포MMP-1기인단백적분비,가입IGFⅡ간예후가감소관절연골세포MMP-1기인단백적분비。결론 IGFⅡ구유현저촉진관절연골세포증식적효과。
Objective To investigate the regulating effects of insulin-like growth factorⅡ( IGFⅡ) on gene expres-sion of matrix metalloproteinases( MMP) -1 in chondrocytes. Methods The condyles of femur chondrocytes of 7 days rats were harvested and cultured with different doses of interleukin( IL) -1β,the monolayer cell cultured of chondrocyte MMP-1 mRNA level were assessed by the enzyme-linked immunosorbent at 24th,48th and 72th hours,harvested and cultured with IGFⅡ. Effect of IGFⅡ was observed. Results IL-1β increased the MMP-1mRNA level respectively in cultures of chondro-cytes,IGFⅡdecreased the MMP-1mRNA level respectively in cultures of chondrocytes. Conclusion IGFⅡ can promote chondrocytes proliferation obviously.
