研究了氯离子、锌离子等对猪精子钙通道的调控作用及其机理。将由健康公猪采得的精液经纱布过滤后分为3组,每组6个重复。第1组精液不加任何物质,第2、3组精液分别加氯化钠液和硫酸锌液,混匀并静置2.5h后,观测精子活力和精子、精清中离子含量与酶活。结果表明:(1)在猪精液中分别加适量的氯离子、锌离子,能刺激精清中钙离子向精子中转移,使得精子内钙离子含量显著地(P〈0.05)增加,精清中钙离子有一定程度的(P〉0.05)减少;(2)加锌或加氯离子,精子活力极显著(P〈0.01)地增强;(3)加锌离子组,精子内碱性磷酸酶、碳酸酐酶、腺苷酸环化酶活性极显著地(P〈0.01)提高,cAMP含量极显著地(P〈0.01)增多;精清内碱性磷酸酶、腺苷酸环化酶活性极显著地(P〈0.01)提高,cAMP含量显著地(P〈0.05)增多;(4)加氯离子组,精清内碱性磷酸酶、腺苷酸环化酶活性极显著地(P〈0.01)提高,精清内cAMP含量显著地(P〈0.05)增多;但精子内碳酸酐酶、腺苷酸环化酶活性和cAMP含量都无显著的(P〉0.05)变化。上述结果表明:猪精液加适量锌离子或氯离子,对精子细胞钙通道功能可能都有上调作用,并能提高精液中碱性磷酸酶、腺苷酸环化酶、碳酸酐酶活性,增加精液中cAMP含量,增强精子活力。
研究瞭氯離子、鋅離子等對豬精子鈣通道的調控作用及其機理。將由健康公豬採得的精液經紗佈過濾後分為3組,每組6箇重複。第1組精液不加任何物質,第2、3組精液分彆加氯化鈉液和硫痠鋅液,混勻併靜置2.5h後,觀測精子活力和精子、精清中離子含量與酶活。結果錶明:(1)在豬精液中分彆加適量的氯離子、鋅離子,能刺激精清中鈣離子嚮精子中轉移,使得精子內鈣離子含量顯著地(P〈0.05)增加,精清中鈣離子有一定程度的(P〉0.05)減少;(2)加鋅或加氯離子,精子活力極顯著(P〈0.01)地增彊;(3)加鋅離子組,精子內堿性燐痠酶、碳痠酐酶、腺苷痠環化酶活性極顯著地(P〈0.01)提高,cAMP含量極顯著地(P〈0.01)增多;精清內堿性燐痠酶、腺苷痠環化酶活性極顯著地(P〈0.01)提高,cAMP含量顯著地(P〈0.05)增多;(4)加氯離子組,精清內堿性燐痠酶、腺苷痠環化酶活性極顯著地(P〈0.01)提高,精清內cAMP含量顯著地(P〈0.05)增多;但精子內碳痠酐酶、腺苷痠環化酶活性和cAMP含量都無顯著的(P〉0.05)變化。上述結果錶明:豬精液加適量鋅離子或氯離子,對精子細胞鈣通道功能可能都有上調作用,併能提高精液中堿性燐痠酶、腺苷痠環化酶、碳痠酐酶活性,增加精液中cAMP含量,增彊精子活力。
연구료록리자、자리자등대저정자개통도적조공작용급기궤리。장유건강공저채득적정액경사포과려후분위3조,매조6개중복。제1조정액불가임하물질,제2、3조정액분별가록화납액화류산자액,혼균병정치2.5h후,관측정자활력화정자、정청중리자함량여매활。결과표명:(1)재저정액중분별가괄량적록리자、자리자,능자격정청중개리자향정자중전이,사득정자내개리자함량현저지(P〈0.05)증가,정청중개리자유일정정도적(P〉0.05)감소;(2)가자혹가록리자,정자활력겁현저(P〈0.01)지증강;(3)가자리자조,정자내감성린산매、탄산항매、선감산배화매활성겁현저지(P〈0.01)제고,cAMP함량겁현저지(P〈0.01)증다;정청내감성린산매、선감산배화매활성겁현저지(P〈0.01)제고,cAMP함량현저지(P〈0.05)증다;(4)가록리자조,정청내감성린산매、선감산배화매활성겁현저지(P〈0.01)제고,정청내cAMP함량현저지(P〈0.05)증다;단정자내탄산항매、선감산배화매활성화cAMP함량도무현저적(P〉0.05)변화。상술결과표명:저정액가괄량자리자혹록리자,대정자세포개통도공능가능도유상조작용,병능제고정액중감성린산매、선감산배화매、탄산항매활성,증가정액중cAMP함량,증강정자활력。
The regulating and controlling roles of chloride and zinc ions to swine sperm calcium channel and their mechanism were studied.Semen from healthy boar were filtered with gauze and divided into three groups,each group having 6 repeats.The group 1 semen was not added anything;the group 2 and 3 semen were added NaCl and ZnSO 4 solution respectively.Being mixed and placed 2.5 h,various parameters of 3 experimental semen were measured.Experimental results showed that:(1) supplementation of optimum chlorine ions or zinc ions in semen could stimulate migration of calcium ions from Seminal plasm to sperm,thus calcium ion concentration in sperm raised significantly(P 〈 0.05),calcium ion concentration in Seminal plasm decreasesd slightly;(2) supplementation of chlorine or zinc ions in semen could strengthen sperm motility highly significantly(P 〈 0.01);(3) after supplementation of zinc ions in semen,the activities of AKP,CA and AC and the concentration of cAMP in sperm increased highly significantly(P 〈 0.01),the activities of AKP and AC in seminal plasm raised highly significantly(P 〈 0.01),the concentration of cAMP in seminal plasm increased significantly(P 〈 0.05);(4) after supplementation of chlorine ions in semen,the activities of AKP and AC in seminal plasm raised highly significantly(P 〈 0.01),the concentration of cAMP in Seminal l plasm increased significantly(P 〉 0.05).These results suggested that supplementation of optimum zinc or chlorine ions in boar semen may up-regulate the function of sperm calcium channel,and can improve the activities of AKP,CA and AC and the concentration of cAMP in semen,strengthen sperm motility.
