柿叶营养丰富,含有维生素C、芦丁、胆碱、黄酮苷、胡萝卜素和多种氨基酸,尤以维生素C含量为多,比一般水果高几倍到几十倍,具有较高的营养价值和医疗保健作用。维生素C又称抗坏血酸,是人体正常生理活动不可缺少的营养成分。测定维生素C的方法有很多种,但这些方法在操作和样品处理上都有不足之处,而高效液相色谱法(HPLC)具有快速、简单、准确度高等特点,更适合用来测定维生素C。本研究以Syncronis C18为分析柱,柱温30℃,进样量10μL,0.25‰的三氟乙酸作为流动相,流速1.0 mL/min,检测波长为243nm进行分析,结果表明,维生素C的出峰时间为5.25 min,应用外标法进行定量分析,维生素C浓度为(50~250)μg/mL范围内与峰面积有良好的线性关系,相关系数达到0.9998,平均加标回收率为101.65%, RSD为0.37%(n=5)。该方法简单快速、精密度高、重现性好,可作为柿叶维生素C的测定一种方法。

Persimmon leaf nutrient is abundant,containing vitamin C, rutin, choline, flavonoid glycosides, carotenoids and amino acids, especially in the vitamin C content is, several times to dozens of times higher than general fruit,so it has high nutritional value and health care function. Vitamin C, also known as ascorbic acid, is essential nutrients for human body normal physiological activities. There are many methods to determintate the vitamin C, but these methods are less than on operation and sample processing,and High performance liquid chromatography (HPLC) is fast, simple, high accuracy, etc,so HPLC is more suitable for the determination of vitamin C. In this study, Syncronis C18 was usded as analytical column at 30 ℃,and the injection volume was 10 μL.The mobile phase was trifluoroacetic acid (0.25‰) with flow rate of 1.0 mL/min and detection wavelength of 243 nm.The retention time of vitamin C was 5.25 min,and external standard method was used to quantitatively detect. The standard working curve of vitamin C had a good linear relation in the concentration range from 50μg/mL to 250μg/mL with peak areas,and the relative coefficient was 0.9998. The average recovery was 101.65%,RSD was 0.37%(n=5)This method is simple and fast, high precision, good reproducibility, and could be used as a method to determinate vitamin C in persimmon leaf.
