目的:探讨心肌肥大过程中有关心肌细胞能量代谢的病理改变以及参附注射液对其的药理作用。方法:提取原代心肌细胞与血管紧张素Ⅱ(AngⅡ)共培养,BCA法检测细胞总蛋白含量、倒置显微镜拍摄并测量细胞直径,高效液相色谱法检测心肌细胞内ATP、ADP、AMP含量并计算腺苷酸含量及能荷水平。在此基础上给予参附注射液,观察参附注射液对心肌细胞能量代谢的药理作用。结果:在心肌细胞肥大过程中,心肌细胞ATP、ADP含量、总腺苷酸含量、能荷水平显著降低(P<0.01),AMP含量显著增加(P<0.01)。参附注射液组较同期模型组,心肌细胞总蛋白含量和心肌细胞直径减少(P<0.05),心肌细胞ATP、ADP含量著提高(P<0.05),AMP含量明显降低(P<0.05),心肌细胞总腺苷酸含量明显升高(P<0.05),心肌细胞能荷水平显著提高(P<0.01)。结论:在心肌肥大过程中,存在能量代谢的紊乱。参附注射液在抑制心肌细胞肥大过程中对能量代谢有益。

목적:탐토심기비대과정중유관심기세포능량대사적병리개변이급삼부주사액대기적약리작용。방법:제취원대심기세포여혈관긴장소Ⅱ(AngⅡ)공배양,BCA법검측세포총단백함량、도치현미경박섭병측량세포직경,고효액상색보법검측심기세포내ATP、ADP、AMP함량병계산선감산함량급능하수평。재차기출상급여삼부주사액,관찰삼부주사액대심기세포능량대사적약리작용。결과:재심기세포비대과정중,심기세포ATP、ADP함량、총선감산함량、능하수평현저강저(P<0.01),AMP함량현저증가(P<0.01)。삼부주사액조교동기모형조,심기세포총단백함량화심기세포직경감소(P<0.05),심기세포ATP、ADP함량저제고(P<0.05),AMP함량명현강저(P<0.05),심기세포총선감산함량명현승고(P<0.05),심기세포능하수평현저제고(P<0.01)。결론:재심기비대과정중,존재능량대사적문란。삼부주사액재억제심기세포비대과정중대능량대사유익。
Objective:To explore pathological changes about energy metabolism during the course of myocar-dial hypertrophy and pharmacological action of ShenFu injection for myocardial cells. Method:Neonatal myocardial cells were abstracted and cultured with AngⅡ, total protein content was detected with BCA method, cellular diame-ter measured with inverted microscope, the contents of ATP, ADP and AMP of myocardial cells by HPLC, the con-tent of adenosine acid and energy charge were calculated. Pharmacological effects of ShenFu injection on energy metabolism were observed when ShenFu injection was administered to myocardial cells. Result:During the process of myocardial hypertrophy, the contents of ATP, ADP and total adenosine acid as well as energy charge were de-creased significantly(P<0.01), the contents of AMP were increased remarkably (P<0.01). The contents of total pro-tein of myocardial cells and cellular diameter were lowered when ShenFu injection group was compared with the model group (P<0.05), the contents of ATP and ADP were significantly increased (P<0.05), AMP contents were lowered obviously (P<0.05), the contents of total adenosine acid were raised remarkably (P<0.05), the level of en-ergy charge were improved notably (P<0.01). Conclusion:There is the disorder of energy metabolism in the process of myocardial hypertrophy. ShenFu injection is helpful to energy metabolism in the course of inhibiting myocardial hypertrophy.
