利用筛选的肠杆菌发酵生产壳聚糖酶,对培养温度、pH和接种量进行优化,确立中心位点后进行响应面设计分析,确定最优发酵培养条件。结果表明:最佳培养工艺条件为培养温度31.74℃、pH 6.5和接种量为7.26%(V/V),培养36 h时壳聚糖酶活性达到13.59 U/mL,实际值与预测值之间的误差约为0.43%。壳聚糖酶的生产方式为延续合成型,具有深入优化并规模生产的潜力。

이용사선적장간균발효생산각취당매,대배양온도、pH화접충량진행우화,학립중심위점후진행향응면설계분석,학정최우발효배양조건。결과표명:최가배양공예조건위배양온도31.74℃、pH 6.5화접충량위7.26%(V/V),배양36 h시각취당매활성체도13.59 U/mL,실제치여예측치지간적오차약위0.43%。각취당매적생산방식위연속합성형,구유심입우화병규모생산적잠력。
Optimization of culture conditions were conducted for chitosanase production by Enterobacter sp.YT21.Firstly Culture temperature , pH and inoculum volume were optimized by one-factor-at-a-time experiments , and then Box-Behnken design and response surface methodology were employed for further optimization of the chitosanase production .The results indicated that the optimal culture conditions were 31.74℃, pH 6.5 and 7.26%(V/V) inoculum, and the highest chitosanase activity obtained was 13.59 U/mL.Error of the actual value compared with the predicted one was only 0.43%, so Enterobacter sp.YT21 was proved to be a suitable potential microorganism for use in chitosanase production .
