Human heart-type fatty acid binding protein ( H-FABP )%Acute myocardial infarction ( AMI)%Colloidal gold immunochromatography technology
目的:应用胶体金免疫层析法制备检测全血或血清样本中心肌型脂肪酸结合蛋白( H-FABP )的检测试纸条,用于急性心肌梗塞( AMI)的早期辅助诊断。方法采用柠檬酸三钠还原法制备胶体金,标记鼠抗心肌型脂肪酸结合蛋白单克隆抗体,喷于玻璃纤维膜上制成胶体金结合物垫,将另一株鼠抗心肌型脂肪酸结合蛋白单克隆抗体和抗鼠二抗分别包被检测线和质控线,组装成试纸条进行灵敏性、特异性、精密性、稳定性及临床样品检测。结果该试纸条的检测灵敏度为10 ng/mL,15 min内可判定结果;与肌钙蛋白I、C反应蛋白、肌酸激酶、人心肌肌红蛋白无交叉反应。检测240份临床标本,与临床诊断结果进行配对分析,阳性符合率95.83%、阴性符合率100%、总符合率97.92%。结论制备的H-FABP检测试纸条有良好的灵敏性、特异性,可用于早期AMI的辅助诊断。
目的:應用膠體金免疫層析法製備檢測全血或血清樣本中心肌型脂肪痠結閤蛋白( H-FABP )的檢測試紙條,用于急性心肌梗塞( AMI)的早期輔助診斷。方法採用檸檬痠三鈉還原法製備膠體金,標記鼠抗心肌型脂肪痠結閤蛋白單剋隆抗體,噴于玻璃纖維膜上製成膠體金結閤物墊,將另一株鼠抗心肌型脂肪痠結閤蛋白單剋隆抗體和抗鼠二抗分彆包被檢測線和質控線,組裝成試紙條進行靈敏性、特異性、精密性、穩定性及臨床樣品檢測。結果該試紙條的檢測靈敏度為10 ng/mL,15 min內可判定結果;與肌鈣蛋白I、C反應蛋白、肌痠激酶、人心肌肌紅蛋白無交扠反應。檢測240份臨床標本,與臨床診斷結果進行配對分析,暘性符閤率95.83%、陰性符閤率100%、總符閤率97.92%。結論製備的H-FABP檢測試紙條有良好的靈敏性、特異性,可用于早期AMI的輔助診斷。
목적:응용효체금면역층석법제비검측전혈혹혈청양본중심기형지방산결합단백( H-FABP )적검측시지조,용우급성심기경새( AMI)적조기보조진단。방법채용저몽산삼납환원법제비효체금,표기서항심기형지방산결합단백단극륭항체,분우파리섬유막상제성효체금결합물점,장령일주서항심기형지방산결합단백단극륭항체화항서이항분별포피검측선화질공선,조장성시지조진행령민성、특이성、정밀성、은정성급림상양품검측。결과해시지조적검측령민도위10 ng/mL,15 min내가판정결과;여기개단백I、C반응단백、기산격매、인심기기홍단백무교차반응。검측240빈림상표본,여림상진단결과진행배대분석,양성부합솔95.83%、음성부합솔100%、총부합솔97.92%。결론제비적H-FABP검측시지조유량호적령민성、특이성,가용우조기AMI적보조진단。
Objective To develop the detection kit of human heart-type fatty acid binding protein ( H-FABP ) from the whole blood or serum samples using colloidal gold immunochromatography technology , for the early diagnosis of acute myo-cardial infarction ( AMI) .Methods The colloidal gold was obtained by trisodium citrate reduction method .Colloidal gold particles labelled with monoclonal antibody of H-FABP were used as the detector reagent and coated on some glass fiber and dried.The other monoclonal antibody of H-FABP and purified anti-mouse IgG were blotted on the test line and control re-gions of nitrocellulose membrane .Then the strip was finished and the detection sensitivity ,specificity, precision,stability were measured ,and clinical samples were done as well .Results The detection sensitivity of this method was 10 ng/mL and the analysis process can be completed within 15 min.There are no cross reactions with cardiac troponin I ,C-reactive protein,human creatine kinase MB and myocardial myoglobin .To compare the agreement of positivity/negativity between clinical samples and results with 240 cases.we found that the positive coincidence rate is 95.83% and negative coinci-dence rate is 100%, and the total compliance rate is 97.92%.Conclusions The test strip for the rapid detection of H-FABP have been made with good sensitivity and specificity , can be used for early diagnosis of AMI .
