目的探讨应用于膜片钳实验的脑片制备的技术和方法。方法新鲜脑组织取材,根据实验需要,取相应脑区组织,振动切片机切片。然后放入孵育槽内孵育备用。后在显微镜下观察细胞的形态结构。结果切片完整,厚薄均匀,脑组织切片细胞活性较高。符合膜片钳实验需求。结论符合实验需求的脑片,从温度控制、取材、切片等环节都要仔细。

Objective To explore the preparation method of the brain slices applying to patch clamp technique.Methods After decol ating a 2-day-old rat,its brain was transferred into a microtome,which cut the brain into slices.Then the brain slices were kept in an incubation chamber for about 1 hour.Final y,the structures of the brain tissue could be observed under a microscope.Results Section complete, with uniform thickness. The cell activity in the brain slices with the method above were higher.The brain slices can meet the needs of patch clamp research.Conclusion A high quality of brain slices, from the control of temperature, selection, slicing step down up.
