腺嘌呤磷酸核糖基转移酶(APRT,adenine phosphoribosyltransferase)是催化腺嘌呤碱基转化为腺嘌呤核苷一磷酸(AMP)的关键酶。运用生物信息学,RT-PCR和RACE方法,从玉米《片中克隆了1个1202 bp的APRT基因cDNA片段,命名为ZmAPT2(GenBank登录号为AY485263)。该cDNA包含1个642 bp的开放阅读框,编码214个氨基酸。RPS-BLAST分析表明,玉米ZmAPT2蛋白有腺嘌呤磷酸核糖基转移酶的催化结构域。推导的玉米ZmAPT2氨基酸与水稻2型、小麦2型、拟南芥2型、大麦的APRT具有较高序列相似性,一致率分别为79%、75%、56%、56%。

Adenine phosphoribosyltransferase(APRT)is an important enzyme for its ability to convert adenine into AMP.The full-length with 1 202 bp cDNA of maize APRT was cloned through a combination of bioinformatics,RT-PCR and RACE,named as ZmAPT(GenBank accession number was AY485263). The cDNA contained an ORF of 642 bp,which coded for a polypeptide of 214 amino acids. RPS-BLAST analysis demonstrated that the maize ZmAPT2 protein contained a catalyzed domain of APRT.The deduced amino acid of ZmAPT2 showed 79%,75%,56%and 56%identity with rice form 2,wheat form 2,A.thalina form 2 and barley respectively.
