目的：比较不同固定剂对过碘酸-希夫氏反应(periodic acid Schif reaction, PAS)显示肝糖原及效果的影响。方法选取成年家兔肝脏，用5种不同固定液作固定剂，不同浓度的过碘酸为氧化剂， PAS整块染色，石蜡切片。观察肝细胞内糖原的显示结果及变化情况。结果①PAS反应为阳性，呈紫红色颗粒，染色均匀，结构清晰；②Bouin氏液和甲醛固定的糖原阳性反应较弱，糖原颗粒稀少、散在；95%酒精、Carnoy氏液、PA-A-F液固定的糖原阳性反应强，效果好，糖原颗粒清晰，染色均匀，细胞形态结构清楚。结论95%酒精、Carnoy氏液、PA-A-F液固定优于Bouin氏液和甲醛固定；PAS染色可以用先染色后进行石蜡包埋的所谓整块染色方法，而且大大优于传统片染的PAS法。
목적：비교불동고정제대과전산-희부씨반응(periodic acid Schif reaction, PAS)현시간당원급효과적영향。방법선취성년가토간장，용5충불동고정액작고정제，불동농도적과전산위양화제， PAS정괴염색，석사절편。관찰간세포내당원적현시결과급변화정황。결과①PAS반응위양성，정자홍색과립，염색균균，결구청석；②Bouin씨액화갑철고정적당원양성반응교약，당원과립희소、산재；95%주정、Carnoy씨액、PA-A-F액고정적당원양성반응강，효과호，당원과립청석，염색균균，세포형태결구청초。결론95%주정、Carnoy씨액、PA-A-F액고정우우Bouin씨액화갑철고정；PAS염색가이용선염색후진행석사포매적소위정괴염색방법，이차대대우우전통편염적PAS법。
Objective to compare the ef ect of the monoblock staining with section staining on the PAS of liver glycogen. Methods Adult rabbit livers were fixed with five dif erent fixative. Livers in experimental group were treated by PAS with monoblock stain to observe the liver glycogen. Livers in control group were treated by PAS with section stain to observe the liver glycogen. Results ① Two methods both showed the liver glycogen in the hepatocytes with positive PAS reaction with purple granules. Compared with section stain, monoblock stain demonstrated the uniform dyeing, clear structures.②Tissue fixed by Bouin solution and formaldehyde showed weak positive reaction of glycogen with sparse and scat ered glycogen particles. Tissue fixed by 95% alcohol, Carnoy's liquid and PA-A-F liquid showed strong positive reaction of glycogen with clear, uniform glycogen particles and clear cellstructures. Conclusions ① The monoblock stain of PAS is superior to the traditional section stain of PAS.②Fixation of 95%alcohol, Carnoy's liquid and PA-A-F liquid is simple, easy to operate with strong specificity and clear positioning.
