目的:研究去苦参碱对HT29细胞的促凋亡活性及其与胞内[Ca2+]i浓度的关系。方法流式细胞术(FCM)及DNAladder法检测细胞凋亡;应用钙荧光指示剂Fluo-3/AM测定细胞内游离Ca2+浓度。结果HT29细胞经苦参碱作用24h后,靶细胞的磷脂酰丝氨酸外化率显著高于对照组,其基因组DNA在琼脂糖凝胶电泳中呈典型的梯状带型;HT29细胞内游离Ca2+浓度进行性升高。结论苦参碱触发的HT29细胞凋亡与胞内游离Ca2+浓度升高密切相关。

목적:연구거고삼감대HT29세포적촉조망활성급기여포내[Ca2+]i농도적관계。방법류식세포술(FCM)급DNAladder법검측세포조망;응용개형광지시제Fluo-3/AM측정세포내유리Ca2+농도。결과HT29세포경고삼감작용24h후,파세포적린지선사안산외화솔현저고우대조조,기기인조DNA재경지당응효전영중정전형적제상대형;HT29세포내유리Ca2+농도진행성승고。결론고삼감촉발적HT29세포조망여포내유리Ca2+농도승고밀절상관。
Objective To study the proapoptotic effect of Matrine on the HT29 Cells and its relationship to the changes of intracellular [Ca2+]i. Methods The phosphatidylserine exposure was measured by Annexin-V binding assay; DNA fragmentation detected by agarose gel electrophoresis; and the effects of Matrine on intracellular [Ca2+]i were studied with Fura 2 microlfuorometry technique. Results After treated with Matrine the phosphatidylserine exposure rate of HT29 were highly increased in Matrine group compared with control, a characteristic DNA“ladder”pattern was found in agarose gel electrophoresis, and the inlfux of extracellularcalcium was progressively elevated. Conclusion Matrine induced apoptosis in HT29 cells, appears to be closely associated with the increase in the concentration of intracellular[Ca2+]i.
