基于脲酶( UASE)催化尿素水解的反应中Cu2+对于酶的抑制作用原理,通过测定产物吸光度的变化率得出了其酶促反应速度,对应于Cu2+浓度而制得标准曲线,检出限为0.05μmol/L.讨论了pH值和温度对测定的影响,结果表明:该法快速灵敏.

A spectroscopic method was developed to determine copper (Ⅱ) content based on the inhibition of copper (Ⅱ) for the hydrolysis of urea catalyzed by urease ( UASE) .The rate of the absorbance change of the product was measured for the calculation of the rate of the enzymatic reaction and a standard curve of copper (Ⅱ) was prepared .The detection limit could reach 0.05μmol/L, and the effects of the pH value and temperature were also discussed .The results indicated that the method was rapid and sensitive .
