以微球菌核酸酶法分离的啤酒酵母(Saccharomyces cerevisiae)染色体单核小体DNA为模板进行PCR,根据不同靶序列PCR产物拷贝数的不同定位核小体位置。结果表明,啤酒酵母Ⅲ号染色体ARS305附近核小体位置大约在39107位到39269位、39318位到39470位及39540位到39694位之间,理论计算结果与试验结果基本一致。
以微毬菌覈痠酶法分離的啤酒酵母(Saccharomyces cerevisiae)染色體單覈小體DNA為模闆進行PCR,根據不同靶序列PCR產物拷貝數的不同定位覈小體位置。結果錶明,啤酒酵母Ⅲ號染色體ARS305附近覈小體位置大約在39107位到39269位、39318位到39470位及39540位到39694位之間,理論計算結果與試驗結果基本一緻。

Using the variation pattern of copy number of PCR product as a function of PCR target sequence, position to the nucleosome locations was determined when isolated mono-nucleosome DNA of Saccharomyces cerevisiae by micrococcal nuclease was used as the PCR template. The results showed that there were three nucleosomes around ARS305 of chromosome Ⅲ located at 39107- 39269 bp, 39318-39470 bp, and 39540- 39694 bp. Computational calculation was in agreement with the experimental data.
