目的:探讨灯盏花素(Br)对脑缺血再灌注大鼠海马核因子(NF)-κB表达的影响。方法采用Zea-Longa法建立脑缺血再灌注动物模型。96只SD大鼠根据不同的处理因素分为:假手术组(Sham组,32只)、灯盏花素治疗组(Br组,32只)、缺血再灌注组(I/R组,32只)。Br组和 I/R组依据缺血后再灌注的不同时间点再细分为3、12、24、48 h 4个小组。应用免疫组织化学方法和蛋白印迹方法检测各组大鼠海马N F-κB蛋白的表达。结果与假手术组相比,在缺血再灌注3 h时,大鼠海马N F-κB的阳性表达已明显升高,在12 h时表达最高,随后表达开始下降(P<0.05);与相同时间点的缺血再灌注组大鼠比较,3 h时,Br组的NF-κB蛋白阳性表达未见明显降低(P>0.05),其余各时间点则明显降低(P<0.05);蛋白印迹方法也得到了相同的结论。结论灯盏花素很可能通过减少NF-κB蛋白的表达改善脑缺血再灌注损伤。

Objective To study the expression of NF-κB in hippocampus of cerebral ischemia-reperfusion rat after breviscapine was injected .Methods The ischemia-reperfusion model was induced with Zea-Longa′s way . Ninty-six SD rats according to the different treatment were randomly divided into the sham group ,ischemia-reper-fusion group(I/R) and breviscapine group(Br) at different time points (3 h ,12 h ,24 h ,48 h) after reperfusion on average .The expression of NF-κB in hippocampus of all groups was examined by immunochemistry and west-ern blot methods .Results Compared with the sham group ,the positive expression of NF-κB in hippocampus of the rats in I/R group was increased significantly at the point of 3 h after reperfusion (P<0.05) ,to the peak at 12 h;compared with the rats in I/R group at the same time point ,the positive expression of NF-κB didn′t evidently reduce at the point of 3 h after breviscapine was injected (P>0.05) ,and decreased significantly in 12 h ,24 h and 48 h groups (P< 0.05);and the same result was obtained by western blot method .Conclusion Breviscapine could down-regulate the expression of NF-κB in hippocampus of cerebral ischemia-reperfusion rat .
