目的：探讨靶向干扰细胞周期调节蛋白依赖性激酶2-关联蛋白-1（CDK2AP1）对乳腺癌MCF-7细胞体外迁移和侵袭的影响。方法构建重组慢病毒 pGCLV- CDK2AP1- shRNA及重组空病毒pGCLV- GFP，分别稳定转染乳腺癌细胞系MCF-7，获得乳腺癌细胞株 MCF-7/RNAi- NC 和 MCF-7/RNAi- CDK2AP1。采用定量 PCR 法检测两组 MCF-7细胞株的CDK2AP1mRNA表达，xCELLigence/RT- CIM实时细胞电子分析系统检测细胞迁移能力，Transwel 实验检测细胞的侵袭能力。结果成功构建乳腺癌细胞株MCF-7/RNAi- NC和MCF-7/RNAi- CDK2AP1，相比MCF-7/RNAi- NC细胞组，MCF-7/RNAi- CDK2AP1细胞组的CDK2AP1mRNA表达受到明显抑制，其细胞迁移和侵袭能力均显著下降（P＜0.05）。结论乳腺癌细胞MCF-7中CDK2AP1低表达能有效抑制细胞的体外迁移和侵袭能力。
목적：탐토파향간우세포주기조절단백의뢰성격매2-관련단백-1（CDK2AP1）대유선암MCF-7세포체외천이화침습적영향。방법구건중조만병독 pGCLV- CDK2AP1- shRNA급중조공병독pGCLV- GFP，분별은정전염유선암세포계MCF-7，획득유선암세포주 MCF-7/RNAi- NC 화 MCF-7/RNAi- CDK2AP1。채용정량 PCR 법검측량조 MCF-7세포주적CDK2AP1mRNA표체，xCELLigence/RT- CIM실시세포전자분석계통검측세포천이능력，Transwel 실험검측세포적침습능력。결과성공구건유선암세포주MCF-7/RNAi- NC화MCF-7/RNAi- CDK2AP1，상비MCF-7/RNAi- NC세포조，MCF-7/RNAi- CDK2AP1세포조적CDK2AP1mRNA표체수도명현억제，기세포천이화침습능력균현저하강（P＜0.05）。결론유선암세포MCF-7중CDK2AP1저표체능유효억제세포적체외천이화침습능력。
Objective To investigate the role of cyclin- dependent kinase 2- associated protein 1(CDK2AP1) in migration and invasion of human breast cancer cells. Methods Recombinant lentivirus pGCLV- CDK2AP1- shRNA and negative control pGCLV- GFP were constructed and transfected to human breast cancer MCF- 7 cells. Migration and invasion ability of MCF- 7 cells were determined with xCELLigence system and Transwel chamber assay, respectively. Results Recombinant lentivirus pGCLV- CDK2AP1- shRNA and negative control pGCLV- GFP were transfected into MCF- 7 cells successful y. After transfection, expression levels of CDK2AP1 mRNA in MCF- 7 cells were reduced significantly(P<0.05). Compared to control MCF- 7/RNAi- NC cells, MCF- 7/RNAi- CDK2AP1 significantly decreased the migration and invasion ability of MCF- 7 cells. Conclusion Si-lence of CDK2AP1 significantly inhibits migration and invasion of human breast cancer MCF- 7 in vitro.
