目的 分离并鉴定夜香树叶的甾体皂苷并测试其抗肿瘤活性.方法 运用反复硅胶柱层析的方法对夜香树叶正丁醇部位进行分离纯化,以LC-MS光谱数据确定其结构;采用MTT法评估分离物对3种肿瘤细胞株的毒性.结果 从夜香树叶的正丁醇部位中分得3个化合物,化合物1为spirost-5-ene-23-diol[2α,3β,25R 3-O-[β-D-glucopyranosyl-(1→3)-β-D-glucopyranosyl(1→2)-β-D-glucopyranosyl{(3→1)-β-D-xylopyranosyl}(1→4)-β-D-galactopyranoside;化合物2为spirost-5-ene-3-diol[3β,25R] 3-O-[{ α-L-rhamnopyranosyl-(1→4)α-L-rhamnopyranosyl-(1-→2)}-α-L-rhamnopyranosyl-(1→4)]-β-D-glucopyranoside;化合物3为spirost-5-ene-23-diol[2α,3β,25R] 3-O-[β-D-glu\opyranosyl-(1→3)-β-D-xylopyranosyl}(1→4)-β-D-galactopyranoside.3种皂苷均可以抑制肿瘤细胞BEL7404、HeLa和A549细胞的生长,并呈现一定剂量依赖关系和时间依赖关系.结论 夜香树甾体皂苷体外对3种肿瘤细胞的抑制作用强度可能取决于鼠李糖苷和吡喃葡萄糖苷的数量与甾体皂苷3号碳上的连接方式.
